大黄对大鼠胃肠道肠球菌属数量的影响 被引量:2

Effect of Rhubarb on the Number of Enterococcus in Gastrointestinal Tract in Rats

摘要 目的:应用实时荧光定量PCR技术检测大黄对大鼠胃肠道肠球菌属数量的影响,为探索大黄对调节肠球菌属数量变化提供参考依据。方法:采用低剂量(0.3g/kg)、中剂量(1.5g/kg)、高剂量(7.5g/kg)大黄水煎剂灌胃干预SD大鼠,并设立空白对照组;在大黄水煎剂灌胃第一、二、三周结束时随机抽取各组动物进行取材,采集大鼠胃、十二指肠、空肠、回肠、结肠各段(含内容物),并提取细菌基因组DNA,采用实时荧光定量PCR法检测各样本不同给药时间下肠球菌属的数量。结果:在中剂量大黄的干预下,胃部一、二、三周肠球菌属数量明显减少(P<0.01);大黄给药一周后,十二指肠各剂量给药组肠球菌属数量明显减少(P<0.01);在空回肠部位大黄给药一周及结肠部位给药二周后,肠球菌属数量有不同程度的减少(P<0.05);而在空肠部位,各剂量给药组在二、三周时肠球菌属数量增多(P<0.05)。结论:大黄水煎剂灌胃能使大鼠胃肠道肠球菌属数量减少,而长时间给药使回肠部位肠球菌属数量增加。 Objective : To analyze the effect of rhubarb on the number of gastrointestinal enterococcus in rats by real-time fluorescence quantitative PCR and to provide reference for rhubarb' s regulating the number of en- terococeus. Methods: SD rats were administered intragastrieally with low dose (0.3/kg) , middle dose ( 1.5/ kg) and high dose (7.5g/kg) rhubarb decoction, while a blank control group was established. SD rats' stomach and segments of their duodenum, jejunum, ileum and colon (including their contents) were gathered at the end of the first, second and third weeks of being administered, and bacterial genome DNA was extracted. Detected the number of enterococcus at different time points of administration by using real-time fluorescent quantitative PCR method. Results:In the middle dose of rhubarb intervention, the number of enterococcus in the stomach significantly reduced (P〈0. 01 ) in the first, second and third weeks. One week after the administe- ring rhubarb, the number of enterococcus in each dose of duodenum decreased significantly (P〈0.01). After administrating rhubarb in the empty ileum for one week and in the colon for two weeks, the number of entero- coccus decreased with varying degrees (P〈0. 05). While in jejunum, the number of enterococcus increased in the second and third week after administering in different dose in every group (P〈0. 05 ). Conclusion: Rhubarb decoction could reduce the number of gastrointestinal enterococcus in rats, and prolonged administration to ileum may increase the number of enterococcus.
