摘要
以采自云南省昆明、迪庆、大理、玉溪、曲靖、楚雄、文山等地的美味牛肝菌子实体为试材,采用ITS、LSU保守区段克隆测序和序列同源性比对的方法,将从GenBank中获得的部分牛肝菌属物种ITS和LSU序列与美味牛肝菌通过邻接法进行系统发育分析。结果表明:基于ITS系列构建的系统发育树表明,美味牛肝菌与Boletus reticulatus和Boletus aereus间序列差异较小,亲缘关系较近,以76%的支持率聚为一簇;基于LSU序列构建的系统发育树表明美味牛肝菌独立成为一枝,且除样品DJ外,其余样品以96%的支持率聚为一簇,表明它们有较近的亲缘关系。
The Boletus edulis fruitbody as the testing material was collected from Kunming,Diqing,Dali,Yuxi,Qujing,Chuxiong,Wenshan and other places in Yunnan Province.The method that employed the cloning sequencing on conserved segment and comparison of sequence homology about ITS and LSU was adopted,which was used to carry out the phylogenetic analysis of part of Boletus edulis ITS and LSU sequences obtained from GenBank.The results indicated that the phylogenetic tree based on ITS sequences showed that the sequence difference of Boletus edulis,Boletus reticulatus and Boletus aereus was smaller and the genetic relationship was closer,clustering with 76% support rate.The phylogenetic tree based on the LSU sequence showed that Boletus edulis became an independent branch,with the exception of sample DJ,the remaining samples clustered with 96%support rate,which indicated that they had a closer genetic relationship.
