摘要
目的:探讨痛泻要方对肠易激综合征肝郁脾虚证大鼠血清、结肠5-羟色胺(5-HT)的调控。方法:将120只SPF级SD大鼠随机分为6组:正常组、病理组、痛泻要方低剂量组、痛泻要方中剂量组、痛泻要方高剂量组、西药(匹维溴铵)组,每组20只。除正常组外,其余各组均用"束缚应激+番泻叶灌胃法"进行造模,前肢束缚2 w,在束缚的基础上进行番泻叶灌胃1 w,1次/d。后用相应剂量的痛泻要方和匹维溴铵进行灌胃治疗7 d。造模及治疗结束后,检测各组大鼠血清及结肠5-HT的含量。结果:与正常组相比,模型组大鼠血清、结肠的5-HT含量显著升高(P<0.01);与模型组相比,痛泻要方低、中、高剂量组和西药组大鼠血清、结肠的5-HT含量明显降低(P<0.05或P<0.01),以痛泻要方高剂量组大鼠的5-HT含量最接近正常组水平。结论:痛泻要方通过疏肝、柔肝的作用,减少大鼠血清和结肠5-HT的含量,来缓解IBS肝郁脾虚证大鼠的症状。
Objective:To discuss effect of Tongxie Yaofang on Serum and Colon 5-serotonin in rats with Liver-depression and Spleen-deficiency irritable bowel syndrome. Method:120 SPF SD rats were divided into six groups with 20 rats in each group:normal group,pathological group,low,middle and high doses group of Tongxie Yaofang and piroxicam group. The model was made by restraint stress and Fanxieye gavage,forelimb restraint 2 weeks except the normal group. On the basis of restraint Sennae intragastric administration 1 weeks for once a day. Western medicine group was given pinaverium bromide,low,middle and high dose groups were given with Tongxie Yaofang of different dose. The contents of 5-HT in serum and colon in each group were detected. Results:Compared with the normal group,the content of 5-HT in the serum and colon of the model group was significantly increased(P〈0.01). Compared with the model group,the contents of 5-HT in serum and colon of Tongxie Yaofang low,medium and high dose groups and the piroxicam group were significantly decreased(P〈0.05,P〈0.01). The content of 5-HT in Tongxie Yaofang high dose group was the closest to that of normal group. Conclusion:Tongxie Yaofang can alleviate the symptoms of IBS with liver-depression and spleen deficiency syndrome by reducing the content of 5-HT in serumard colon.
