摘要
【摘要】目的人类TMEM121基因是一个在心脏的胚胎和成体期均有高表达的基因,尽管该基因已被克隆十余年,但其生物学功能迄今仍是一片空白。方法本研究利用生物信息学方法和工具对人类TMEM121蛋白的理化性质、二级结构、亲/疏水性、互作蛋白及功能等进行预测分析。结果显示TMEM121蛋白在进化上高度保守,与猩猩、绿猴、家猫、大鼠及小鼠等的同源度高达96%以上,d螺旋是构成该蛋白二级结构的主要元件,无信号肽,定位于内质网的可能性比较大,具有转运及信号转导等功能。结论人类TMEM121基因可能作为转运和信号转导因子与其他蛋白共同作用参与成体心脏功能的调控。
Objective Human TMEM12I gene, a highly expressed gene in both fetal and adult heart, has been identified for decades, but it' s in vivo biological function remains unknown. Methods To analyze the structure and function of TMEM121 protein with bioinformatics, the bioinformatic methods and tools were used to analyze the physical and chemical properties, secondary structure and functional category of TMEM121 protein. Results The bioinformatic analysis revealed that the proline content of TMEM121 protein was highly conserved in evolution, the main secondary structure element was alpha helix without sig- nal peptide and etc. The TMEM121 protein were mainly localized in endoplasmic reticulum and played a role in transporter regulation and signal transducer. Conclusions Human TMEM121 gene may act as signal transducer factor participating in adult cardiac function.
