摘要
膜片钳技术是生命科学研究中的重要技术手段,利用膜片钳技术可以准确地记录到细胞膜上离子通道的电活动和囊泡的分泌特性。在神经科学方面,膜片钳技术有助于阐明神经元内部蛋白分子的生理活动和功能,探究相关疾病的分子机制并提供潜在的治疗方法。但是,膜片钳实验的操作却有一定难度,初学者需要经过系统学习和长时间练习才能很好地掌握操作方法。本文以最常见的电压钳全细胞模式为例,结合膜片钳的具体操作过程,陈述了实验操作中应该特别注意的一些细节和问题,以协助实验人员记录到有效的实验数据。
Patch-clamp technique plays an important role in life science research, which can accurately record the electrical activity of ion channels on cell membrane and vesicle secretion characteristics. In neuroscience, patch-clamp technique helps to elucidate the physiological activity and function of protein molecules in neurons, explore the molecular mechanism of related diseases and provide potential therapeutic methods. However, the operation of patch-clamp experiment is difficult and beginners need to learn and practice for a long time to master the operation method. In this paper, the voltage-clamp wholecell model were presented as an example, and according to the specific operation process of patch-clamp, some details and problems which should be paid special attention to in the experimental operations were stated, and this can help experimental operators get effective experimental data.
