加味五子衍宗合剂及组分对生精细胞损伤模型大鼠生殖激素水平的影响 被引量:8

Effects of flavored Wuzi Yanzong mixture and its components on reproductive hormone levels of spermatogenic cells damaged rat model

摘要 目的探讨加味五子衍宗合剂及其组分中药对腺嘌呤所致生精细胞损伤模型大鼠生殖激素水平的影响。方法雄性SD大鼠80只,10只作为正常组,70只大鼠按20 mg·100 g^(-1)体重灌胃给予腺嘌呤,每日1次,连续4周,制备大鼠生精细胞损伤的少弱精模型,将造模成功的60只大鼠随机分为模型组、加味五子衍宗合剂组、五子衍宗方组、仙茅+淫羊藿水提物组、仙茅水提物组、淫羊藿水提物组,每组10只。造模成功后,各给药组按6 mL·kg^(-1)体重灌胃给药(浓度分别为432、180、72、24、48 mg·mL^(-1)),每日1次,连续6周,正常组、模型组每日灌胃给等体积的生理盐水。观察大鼠精子数、精子活动率和精子指数及血清睾酮(T)、卵泡刺激素(FSH)和黄体生成素(LH)改变。结果与正常组比较,模型组大鼠精子数、精子活动率及精子指数均显著下降(P<0.01),血清T含量降低(P<0.05),LH含量显著升高(P<0.01),血清FSH含量有升高趋势,但无显著差异(P>0.05)。与模型组比较,各给药治疗组大鼠精子数、精子活动率及精子指数显著升高(P<0.01);加味五子衍宗合剂组显著多于其他组分组(P<0.01),与五子衍宗方组作用相近(P>0.05)。与模型组相比,除淫羊藿水提物组外,其余各组大鼠血清T含量升高、LH含量降低(P<0.05或P<0.01),且加味五子衍宗合剂组显著优于其他组分组(P<0.05)。结论加味五子衍宗合剂可调节生精细胞损伤模型大鼠生殖激素T和LH水平,改善精子数和活动率。 AIM To investigate the effects of flavored Wuzi Yanzong mixture and its components on reproductive hormone levels of the spermatogenic cells damaged rat model induced by adenine. METHODS Eighty male SD rats, ten of which as normal group. The model of the rare and weak cell injury of the rats was prepared of adenine 20 mg. 100 g-1 per day with gavage for 4 weeks. Sixty model rats were randomly divided into model group, flavored Wu zi Yanzong mixture group, Wuzi Yanzong prescription group, water extract from curculigo-epimedium group, water extract from curculigo group and water extract from epimedium group, 10 rats in each group. Rats in each group were given with gavage at a dose of 6 mL.kg-1 (432, 180, 72, 24, 48 mg.mL-1, respectively) once a day for 6 weeks, and the normal group and model group were given an equal volume of normal saline. Sperm count, sperm activity rate and sperm index of rats were observed. Serum testosterone (T), follicle- stimulating hormone (FSH), and luteinizing hormone (LH) were detected. RESULTS Compared with the normal group, sperm count, sperm activity rate and sperm index of the model group decreased significantly (P 〈 0.01), serum T decreased (P 〈 0.05), LH content increased significantly (P 〈 0.01) FSH content hadincreased trend with no significant difference (P 〉 0.05). Compared with the model group, sperm count, sperm activity rate and sperm index of rats in each treatment groups were significantly increased (P 〈 0.01), which in the flavored Wuzi Yangzong mixture group was significantly more than those in the other components groups (P 〈 0.01) and similar with the Wuzi Yanzuan prescription group (P 〉 0.05). The contents of serum T and LH in other groups significantly lower than those in the model group (P 〈 0.05 or P 〈 0.01) , except for the water extract from epimedium group. Moreover, which in the flavored Wuzi Yanzong mixture group was significantly better than those in the other components groups (P 〈 0.05). CONCLUSION Flavored Wuzi Yanzong mixture can regulate the levels of serum T and LH in spermatogenic cells injury model rats and improve the sperm count and activity rate.
