运动干预对糖尿病前期人群KCNJ11基因多态性与糖脂代谢关系的影响 被引量:3

Effects of exercise intervention on the relationship between KCNJ11 gene polymorphism and glucose and lipid metabolism in pre-diabetes mellitus

摘要 目的分析运动干预对糖尿病前期人群的钾离子内向整流通道蛋白亚单位11(KCNJ11)基因多态性与糖脂代谢关系的影响。方法于2017年1—3月采用分层整群随机抽样方法,对山东聊城市5个社区46例糖尿病前期人群进行8周的运动干预,在运动干预前后进行KCNJ11基因多态性分析与糖脂代谢指标检测,并探讨两者之间的相关性。结果与运动干预前相比较,运动干预8周后,46例糖尿病前期人群的空腹血糖、餐后2 h血糖、糖化血红蛋白及血脂均明显改善(P<0.05)。KCNJ11基因多态性分析可见,运动干预后rs5219位点的CC基因型人群的甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)脂代谢指标变化较TT、CT基因型更显著(t=2.635、3.213和2.048,P=0.012、0.003和0.048),运动干预后rs2285676位点的GG基因型人群的空腹血糖、糖化血红蛋白糖代谢指标变化较AA、AG基因型更显著(t=2.236和2.062,P=0.034和0.049)。结论 KCNJ11基因rs5219位点的CC基因型与rs2285676位点的GG基因型,可作为糖尿病前期人群运动干预时评价糖脂代谢指标变化的优势基因。 Objective To investigate the effect of exercise intervention on the relationship between KCNJ11 gene polymorphism and glycolipid metabolism in pre-diabetic population. Methods From January 2017 to March 2017, 5 communities were selected by stratified cluster random sampling method in Liaocheng City, Shandong Province, and 46 cases of pre-diabetes' data were collected through public welfare physical examination activities The exercise intervention was performed for 8 weeks with their consent and cooperation. The polymorphism of KCNJll gene and the index of glucose and lipid metabolism were detected before and after exercise intervention, and the correlation between them was also discussed. Results After 8 weeks of exercise intervention, 2 h postprandial blood glucose, fasting glucose, glycosylated hemoglobin and four indicators of blood lipids were significantly improved in pre-diabetes populations (P 〈 0. 05). KCNJ11 gene polymorphism analysis showedthat the TG, TC and HDL-C lipid metabolism indexes of CC genotype in rs5219 locus were more significant than those of TF and CT genotypes (t = 2. 635,3. 213 and 2. 048 ,P = 0. 012, 0. 003 and 0. 048 ) after exercise intervention, the changes of FPG and HbAlc glucose metabolism indexes in GG genotype population of rs2285676 locus were more significant than those in AA and AG genotypes (t = 2. 236 and 2. 062,P = 0. 034 and 0. 049). Conclusion The CC genotype of the KCNJll gene rs5219 site and the GG genotype of rs2285676 site can be used as the dominant gene to evaluate the change of glucose lipid metabolism in the early stage of diabetes intervention.
