白芥子挥发油促进冬病夏治方经皮渗透研究

Effect of Volatile Oil from Sinapis Semen on Percutaneous Permeability of the “Treatment of Winter Disease in Summer” Prescription

目的研究白芥子挥发油对冬病夏治方中多个有效成分的促进透皮吸收作用。方法采用体外透皮试验,以离体大鼠皮肤为透皮屏障,以冬病夏治方各中药中的有效成分延胡索乙素、细辛脂素、盐酸麻黄碱、黄芩苷作为模型药物,HPLC法测定各模型药物透过量,观察白芥子挥发油(0.5%、2%、5%)对各模型药物的经皮促渗透作用。结果白芥子挥发油浓度较低时(0.5%)对各模型药物无明显的促进经皮渗透作用,随着浓度的增大,白芥子挥发油(2%,5%)可显著促进有效成分延胡索乙素、细辛脂素、盐酸麻黄碱、黄芩苷经皮渗透吸收,且呈浓度依赖性,即浓度越大,促透效果越好。结论白芥子挥发油能促进冬病夏治方中有效成分延胡索乙素、细辛脂素、盐酸麻黄碱、黄芩苷经皮渗透吸收,白芥子作为处方中君药起到了治疗与促进药物渗透双重作用,从而达到增效的目的。

Objective To study the effects of volatile oil from Semen sinapis on promoting the transdermal absorption of multiple effective ingredients in ＂treatment of winter disease in summer＂ prescription. Methods Using rat skin as transdermal barrier and the components of ephedrine hydrochloride,asarinin,tetrahydropalmatine and baicalin as model drugs,the in vitro transdermal test was used to investigate the effect of different concentration of volatile oil from Sinapis semen（0.5%,2%,5%） on the transdermal permeation to each model drug. The HPLC method was used to measure the permeation quantity of model drugs. Results Low concentration（0.5%） of volatile oil from Semen sinapis had less obvious effect on the percutaneous permeation of the model drugs. With the concentration increased, the volatile oil significantly promoted percutaneous penetration of the active ingredients including tetrahydropalmatine, asarinin, ephedrine hydrochloride, baicalin in a concentration-dependent manner. That is,the higher concentration,the better effect of promoting penetration. Conclusion The volatile oil of Semen sinapis can promote the percutaneous absorption of ephedrine hydrochloride, asarinin, tetrahydropalmatine and baicalin,which are the active ingredients in the ＂treatment of winter disease in summer＂ prescription. As the monarch drug in the prescription,it played a dual role in curing and promoting drug permeation of other drugs.
