复方丹参滴丸处理对大鼠移植心脏心肌细胞保护作用及细胞凋亡线粒体途径的影响 被引量:2

摘要 目的:观察复方丹参滴丸处理对大鼠移植心脏心肌细胞保护作用及细胞凋亡线粒体途径的影响。方法:SD大鼠48只随机分为4组:(1)单纯移植组,术前未用任何药物。(2)缺血后处理组:术前未用任何药物,移植术毕,供心升主动脉完全开放前间断开放15s阻断15s,共3次。(3)复方丹参滴丸处理组,供体每日灌胃1次,连续7天,复方丹参滴丸溶于5m L生理盐水中。(4)模型对照组,供体于相应时间内等量生理盐水灌胃,每日1次,连续7天。实验结束后24h,分别处死各组动物取其心肌组织,利用Western blot对P53、Bcl-2及Bax蛋白水平进行分析,再利用TUNEL染色法及Western blot,对心肌细胞凋亡情况进行分析。结果:相较于其他3组,复方丹参滴丸组大鼠心肌细胞凋亡指数及Bax、P53蛋白阳性染色指数、Bcl-2的蛋白阳性表达较稳定,差异有统计学意义(P<0.05);单纯移植组、缺血后处理组以及模型对照组3组间各项指标比较差异无统计学意义(P>0.05)。结论:复方丹参滴丸对大鼠移植心脏缺血再灌注损伤具有保护作用,可能与其通过p53调控Bax和Bcl2的表达水平抑制心肌细胞凋亡机制有关。 Objective : To observe the protective effect of Compound Salvia Dripping Pills on cardiomyocyte apoptosis and mitochondrial pathway in rat cardiac allografts. Method : 48 Sprague-Dawley rats were randomly divided into 4 groups : (1)transplantation group, without any medication before surgery, (2)ischemic postconditioning group: without any medication before surgery, ischemic postconditioning induced by 3 conditioning cycles of 15s of reperfusion and ischemia before complete reperfusion to the donor heart, (3) Compound Salvia Dripping Pills treatment group, daily gavage for 7 days, Compound Salvia Dripping Pills dissolved in 5mL saline,(4) model control group, at the same time the same amount of saline garage once a day for 7 days. Twenty-four hours after the experiment, each group of animals were sacrificed and the myocardial tissue was harvested. The protein levels of P53, Bcl-2 and Bax were analyzed by Western blot. TUNEL staining and Western blot was used to analysis myocardial apoptosis condition. Result: Compared with that of the other three groups, the myocardial apoptosis index, Bax, P53 protein positive staining index, the Bcl-2 protein positive expression of Compound Salvia Dripping Pills group was relatively stable, with significant difference ( P〈0.05 ) . There was no significant difference among the three groups in the simple transplantation group, the ischemic posttreatment gronp and the model control group (P〉0.05) .Conclusion: Compound Salvia Dripping Pills have protective effect on cardiomyocytes'in rat cardiac allograft, which has possible relationship with inhibition of the cardiomyocytes apoptosis by regulating the expression level of Bax and Bcl-2 through p53.
