摘要
目的观察复方贞术调脂胶囊(FTZ)对体外培养破骨细胞的影响,探求相关作用机制。方法采用体外分离培养破骨细胞,随机分为control组、Alendronate组、FTZ-H组、FTZ-M组和FTZ-L组,各组大鼠予含药血清、M-CSF和RANKL细胞刺激因子诱导培养8 d,电子显微镜下观察破骨细胞数量和骨陷窝面积、RT-PCR法检测Trap、MMP-9、NFATc1 mRNA的表达。结果经Trap染色,均可在5倍镜下见大小不一、细胞核数不一的多核破骨细胞,与control组相比,Alendronate组、FTZ-H组和FTZ-M组的破骨细胞(OC)数量减少(P<0.05,P<0.01),FTZ组间比较,FTZ-H组较FTZ-L组破骨细胞数量显著降低(P<0.05);Alendronate组、FTZ-H组和FTZ-M组的骨陷窝面积较control组显著减少(P<0.05),FTZ组间比较,FTZ-H组骨陷窝面积较FTZ-L组减少,但差异无统计学意义(P>0.05)。与control组比较,Alendronate组、FTZ-H组Trap、MMP-9、NFATc1 mRNA表达均显著降低(P<0.01)。结论 FTZ-H和FTZ-M浓度具有抑制破骨细胞分化和骨吸收活性的作用,其中FTZ-H组与Alendronate组无异。
Objective To investigate the effect of FTZ on the bone absorption ability of osteoclasts in vitro. Methods Osteoclasts were separated from C57 B6/J of 4 -6w mice and cultured. They were randomly divided into five groups, control, Alendronate, FTZ - H, FFZ - M and FFZ - L groups. After 8 days of cultivation, the number of osteo- clast and bone absorptive lacunae were counted by electron microscope. Trap, MMP -9 and NFATcl mRNA were as- sessed by RT - PCR. Results Compared with the control group, the number of osteoclasts and the area of bone absorptive lacunae were significantly reduced Alendronate, FTZ - H and FTZ - M groups ( P 〈 0. 01, P 〈 0.05 ). Compared with FFZ - L group, significant reduction in the number of osteoclasts was observed in FTZ - H group. There is no significant difference between FFZ - L and FTZ - H in the area of bone absorptive lacunae ( P 〉 0. 05 ). The gene expressions of Trap, MMP- 9 and NFATcl were significantly lower in Alendronate and FTZ -H groups than that in control group (P 〈 0. 01 ). Conclusion FTZ - H and FTZ - M inhibit osteoblast differentiation and bone absorbing activity, but there is no significant difference between FTZ - H and FTZ - M.
