摘要
目的探讨模拟低氧环境下大鼠视网膜新生血管形成的可行性,并探究VEGF-B与视网膜新生血管之间的相关性。方法低氧干预大鼠(置于低压氧舱7d)在进舱前一天在玻璃体腔分别注射抗VEGF-B抗体[低氧药物组,0.2%(W/V),50μg/kg]和同等体积的溶媒(低氧药物对照组,50%PBS+50%甘油)。每组取5只大鼠行视网膜FITC-dextran铺片,剩余7只大鼠采用ELISA法考察玻璃体VEGF-B含量、RT-qPCR法考察视网膜VEGF-B mRNA表达水平。结果视网膜FITC-dextran铺片显示低氧环境下大鼠视网膜中央血管部分丢失并出现新生血管丛,可见血管周围荧光素渗漏。低氧空白组和低氧药物对照组大鼠视网膜VEGF-B mRNA的表达高于正常对照组与低氧药物组(P<0.05),玻璃体VEGF-B的含量在各组中的变化趋势与其相似。结论低氧环境可诱导视网膜新生血管形成,VEGF-B在低氧诱导视网膜新生血管形成过程中发挥了促进作用,而经抗VEGF-B抗体干预后可有效抑制这一作用。
Objective To explore the feasibility of retinal neovaseularization in rats with simulated hypoxia, and explore the correlation between VEGF-B and retinal neovascularization.Method Hypoxic intervention rats (in hypo- baric chamber 7d)were injected with anti VEGF-B antibody[ hypoxia drug group, 0.2% (W/V), 50μg/kg] and the same volume of solvent( hypoxia drug control group ,50%PBS+50% glycerol)in the vitreous cavity a day before the hypoxie intervention.In each group, 5 rats were taken for retinal FITC-dextran paving.The remaining 7 rats were ex- amined by ELISA method to investigate the content of VEGF-B in the vitreous body, and the expression level of VEGF-B mRNA in retina were detected by RT-qPCR.Result The retinal FITC-dextran paving showed that the central vascular part of the retina of rats was lost and the new vascular plexus appeared in the hypoxic environment, and the fluorescein percolation was seen around the blood vessels.The expression of VEGF-B mRNA in hypoxic blank group and hypoxic control group was higher than that in normal control group and hypoxia group(P〈O.05 ) ,and vitreous VEGF-B content in each group had similar trend.Conclusion Hypoxic environment can induce retinal neovascularization.VEGF-B plays a promoting role in hypoxic induced retinal neovascularization, and it can be ef- fectively inhibited by intervention with anti VEGF-B antibody.
