藏药雪灵芝粗多糖对S180荷瘤小鼠肿瘤生长的影响

Effect of Crude Polysaccharide from Tibetan medicine Arenaria kansuensis on Tumor Growth in Sarcoma 180 Tumor-bearing Mice

目的观察藏药雪灵芝粗多糖对S180荷瘤小鼠肿瘤生长的影响。方法建立S180荷瘤小鼠模型,将其随机分为对照组,环磷酰胺(CTX)组,AKCP高(AKCP-H)、中(AKCP-M)、低剂量(AKCP-L)组,连续给药10 d后处死,取瘤称重并计算抑瘤率;HE染色法观察瘤组织形态;荧光定量PCR及Western blot法检测瘤组织中增殖相关因子Cyclin D1、PCNA、Ki67,凋亡相关因子Bcl-2、Bax的m RNA和蛋白的表达。结果 CTX、AKCP-H、AKCP-M、AKCP-L组瘤体质量低于对照组(P<0.05),抑瘤率分别为58.033%、25.574%、31.803%、23.279%;HE染色显示,CTX组及AKCP各剂量组肿瘤组织出现不同程度坏死区及间质增多现象;与对照组比较,各实验组Cyclin D1、PCNA、Ki67、Bcl-2、Bax、Bcl-2/Bax的m RNA及蛋白表达无显著差异(P>0.05)。结论雪灵芝粗多糖在一定程度上抑制了S180荷瘤小鼠肿瘤生长,可能具有抑瘤作用。

Objective To investigate the effects of Tibetan medicine Arenaria kansuensis crude polysaccharide （AKCP）on tumor growth in Sarcoma 180 tumor-bearing mice.Methods Balb/c mice inoculated with S180 cells were randomly divided into five groups：control, CTX, AKCP-H, AKCP-M and AKCP-L groups.For 10 days of drug administration,mice were sacrificed and dissected.The tumors were weighed ,and the tumor inhibition rate was cal- culated.HE staining was used to observe the morphology of the tumor.Expressions of proliferation factors（ CyclinD1, PCNA ,Ki67）and apoptosis factors（Bax,Bcl-2） were detected by quantitative real-time PCR and western blot.Re- sult The tumor weight in CTX,AKCP-H , AKCP-M and AKCP-L groups were less than control group（ P〈0.05）, and in these groups ,the tumor inhibition rate were 58.033% ,25.574% ,31.803% and 23.279%.HE staining showed that in the tumor tissues of the CTX group and three AKCP groups, there were varying degrees of necrosis, the num- ber and the distribution of tumor cells were decreased and sparse, the interstitial mass increased and the leukocytesinfiltrated.There were no difference of mRNA and protein expression levels of CyclinD1, PCNA, Ki67, Bcl-2, Bax and Bcl-2/Bax among control group and other groups （P〉0.05）.Conelusion AKCP may have antitumor effect that inhibits the tumor growth of S180 tumor-bearing mice to a certain extent.
