补肾方药含药血清对UMR106细胞增殖及碱性磷酸酶活性影响

Effects of Medicated Serum of Tonifying Kidney Prescription on Proliferation and Alkaline Phosphatase Activity of UMR106 Cells

目的：研究补肾方药含药血清对UMR106细胞增殖及碱性磷酸酶（ALP）活性的影响。方法：将实验动物随机分成空白血清组、低剂量血清组、中剂量血清组、高剂量血清组、阳性对照组,予以分别灌胃制备含药血清。运用CCK8法测定空白血清组、低剂量血清组、中剂量血清组、高剂量血清组、阳性对照组对UMR106细胞24、48、72 h增殖情况的影响;并测定UMR106细胞24、48、72 h的ALP活性情况。结果：各组自身比较后,加药干预24、48、72 h后各组细胞吸光度值与ALP活性均有增加,差异有统计学意义（P〈0.05）;各组与空白血清组比较,中、高剂量血清组和阳性对照组加药干预24、48、72 h后,细胞吸光度值之间均较低,差异有统计学意义（P〈0.05）,而低剂量血清组与空白血清组分别加药干预24、48、72 h的吸光度值比较均无显著差异（P〉0.05）。与空白血清组比较,低、中、高剂量血清组和阳性对照组加药干预24、48、72 h后,细胞ALP活性均较高,差异有统计学意义（P〈0.05）,且剂量浓度越高,ALP活性越高。结论：表明补肾方药（骨康方）含药血清有促进UMR106细胞增殖及ALP活性的作用。随着补肾方药（骨康方）浓度的增高,其含药血清促进UMR106细胞增殖作用减弱,但ALP活性随着浓度增高而增高。

Objective：To study the effect of medicated serum of tonifying kidney prescription on proliferation and alkaline phosphatase（ALP）activity of UMR106 cells. Methods：The experimental animals were randomly divided into blank serum group,low dose serum group,medium dose serum group,high dose serum group,and positive control group. Respectively intragastric administration preparation containing serum,and the cells were cultured. The proliferation of UMR106 cells and the activity of ALP after 24,48,72 hours of culture. Results：The self comparison after dosing intervention 24 h,48 h,72 h cell proliferation and ALP activity of each group was increased,the difference was statistically significant（P〈0.05）;compared with the blank serum group,medium and high dose drug serum group and positive control group intervention 24 h,48 h,72 h,between the absorbance of cells yields are low,the difference was statistically significant（P〈0.05）,high dose and low absorbance colonization serum group and blank serum group were 24 h and 48 h dosing intervention,72 h showed no significant difference（P〈0.05）. Compared with the blank serum group,the ALP activity of the low,medium and high dose serum group and positive control group after intervention of 24 h,48 h and 72 h were higher,the difference was statistically significant（P〈0.05）,and the higher the dose concentration,the higher the activity of ALP. Conclusion：The results suggest that the prescription（Gukang Decoction）medicated serum on promoting the proliferation of UMR 106 cells and ALP activity. With the prescription（Gukang Decoction）concentration increased,the weakening of the effect of UMR106 containing serum promote cell proliferation,but the activity of ALP increased with the concentration.
