溴氰菊酯暴露对白纹伊蚊抗性株幼虫氨基酸水平的影响 被引量:4

摘要 目的观察溴氰菊酯暴露对抗性白纹伊蚊幼虫氨基酸的影响,探索溴氰菊酯毒性相关氨基酸标志物。方法从广州市越秀区采集白纹伊蚊幼虫,实验室饲养,取F1代幼虫进行溴氰菊酯抗性测定;将抗性白纹伊蚊F1代幼虫分成药物暴露组、丙酮暴露组和空白对照组(每组10条),前两组分别用溴氰菊酯丙酮溶液和相同浓度的丙酮暴露,空白对照组不做处理;用LC-MS/MS检测三组幼虫体内的氨基酸水平,每组重复检测6次。结果 F1代幼虫LC_(50)=0.065 mg/L,确定为抗性种群;药物组幼虫体内12种氨基酸与空白对照组出现差异(P<0.05),其中丙氨酸、瓜氨酸、酪氨酸、甘氨酸、亮氨酸、鸟氨酸、苯丙氨酸、脯氨酸、丝氨酸、蛋氨酸和苏氨酸11种氨基酸水平在暴露后降低(P<0.05),组氨酸水平在暴露后升高(P<0.05);药物组幼虫体内的丙氨酸、瓜氨酸、亮氨酸和苯丙氨酸的变化与丙酮暴露组比较差异有统计学意义(P<0.05)。结论溴氰菊酯丙酮溶液暴露可导致抗性白纹伊蚊幼虫体内12种氨基酸水平改变,其中丙氨酸、亮氨酸、苯丙氨酸和瓜氨酸浓度水平降低与溴氰菊酯暴露有关。 Objective To observe the changes of amino acid level in Aedes albopictus larvae under deltamethrin exposure and explore the biomarkers related to the dehamethrin toxic. Methods The Aedes albopictus larvae were collected from the Yuexiu district of Guangzhou city where was reported that the mosquitoes were highly resistant to deltamethrin. The F 1 generation larvae were collected and determined deltamethrin resistance. The F1 generation larvae were divided into three groups: detalmethrin exposure group (dissolved in acetone), acetone exposure group and blank control group, 10 larvae in each group. The amino acids level in 3 groups were detected by LC-MS/MS, and 6 times repeated detection were completed for each group. Results The LCs0 of the F1 generation larvae of the wild strain Aedes albopictus was 0.065 mg/L, and it can be identified as resistant strain according to WHO criteria (more than 5 times). The levels of 12 out of the 17 common amino acids showed changes in the larvae after drug exposure (P〈0.05). The levels of alanine, citrulline, tyrosine, glycine, leucine, ornithine, phenylalanine, proline, serine, methionine and threonine were decreased after exposure (P〈0.05) , while the level of histidine was increased after exposure (P〈0.05). The change of alanine, citrulline, leucine and phenylalanine levels in the larvae of the detalmethrin exposure group was significantly different from those of the acetone exposure group (P〈0.05). Conclusions Both deltamethrin and acetone might have effects on the levels of some amino acids. Subtracting the acetone effect, dehamethrin exposure could alter the levels of alanine, leucine, phenylalanine and citrulline in the wild deltamethrin-resistant strain of Aedes albopictus larvae.
