摘要
通过对样品进行酶解法前处理,检测结果进行折算,建立了利用高效液相色谱蒸发光散射检测器(HPLC-ELSD)测定婴幼儿配方乳粉中低聚果糖总量的方法。实验优化了酶的加入量、色谱柱、流动相比例、雾化气流量等条件。结果表明:三水平的加标回收率均在86.7%~105.1%之间;相对标准偏差小于5%,检出限为1.0 g/kg。5批次实际样品检测结果和国标方法对比,绝对偏差均小于8%。试验所需仪器检测机构普遍配备,与国标方法相比具有更高的实用性和推广性。
A method for the determination of the total amount of fructo-ohgosaccharide in infant formula milk powder was established for the first time by high performance liquid chromatography with evaporative light scattering detector (HPLC - ELSD). The sample was pro- cessed by enzymatic hydrolysis, and the test result was converted. The test optimizes the conditions of enzyme addition, chromatographic column, mobile phase ratio and flow rate of atomization gas. The results indicated that the recovery of the three levels was between 86.7%- 105.1%. The relative standard deviation is less than 5%, and the detection limit is 1.0 g/kg. Compared with the national standard method, the absolute deviation is less than 8%. The instrument is more practicable and more effective than the national standard method.
