摘要
目的探讨白藜芦醇(Res)对PM2.5诱导气道上皮细胞IL-8表达的影响。方法先用不同浓度PM2.5刺激人气道上皮细胞株16HBE,再将16HBE细胞分为PM2.5组(75 mg/L)、Res+PM2.5组(100μmol/L Res+75 mg/L PM2.5)、空白对照组,分别用荧光定量PCR、ELISA检测表达。结果IL-8 PM2.5呈浓度和时间依赖性刺激16HBE细胞表达IL-8,Res+PM2.5组中IL-8表达明显低于PM2.5组(P<0.05)Res。结论可能通过抑制气道上皮细胞表达IL-8,进而减轻PM2.5诱导的气道炎症反应。
Objective To investigate the effect of resveratrol on IL-8 expression of airway epithelial cells induced by PM2.5.Methods 16HBE cells(human airway epithelial cell line)were stimulated with different concentrations of PM2.5,and then were divided into 3 groups:PM2.5 group(75 mg/L),resveratrol+PM2.5 group(100μmol/L resveratrol+75 mg/L PM2.5),and blank control group.IL-8 expression was detected by fluorescent quatititive PCR and ELISA.Results PM2.5induced IL-8 expression of 16HBE cells in a dose and time dependent manner.IL-8 expression was lower in resveratrol+PM2.5 group than in PM2.5 group(P〈0.05).Conclusion Resveratrol may alleviate PM2.5-induced airway inflammatory reaction via inhibiting IL-8 expression of epithelial cells.
