猕猴桃属33份核心雄性种质SSR指纹图谱构建 被引量:5

摘要 通过对33份猕猴桃核心雄性种质材料进行指纹图谱构建及遗传多样性分析,可为猕猴桃品种鉴定及选育工作提供帮助,也为今后规范进行知识产权的保护提供科学的理论依据。从猕猴桃SSR数据库中选取100对SSR引物,利用遗传背景差异大的4份猕猴桃雄性种质材料进行筛选,选出14对扩增条带清晰、具高多态性且重复性好的引物,对本研究供试的33份猕猴桃核心雄性材料进行扩增,利用8%聚丙烯酰胺凝胶对PCR扩增产物进行检测。筛选得到的14对SSR引物共扩增出171条条带,其中含有163条多态性条带,占95.32%。最少可以通过5对高多态性SSR引物即能将33份猕猴桃雄性种质材料全部区分开。这5对SSR引物共扩增出83条条带,多态性条带82条,占98.80%,每个位点的等位基因为1~12个,平均每对引物为16.6个。通过对33份猕猴桃雄性种质资源进行鉴定,发掘出猕猴桃雄性种质优异标记位点UDK96-035、UDK96-053、UDK96-040、Thr801、For13,可为后续相关研究提供坚实的理论基础。 The fingerprint construction and the analysis of genetic diversity about 33 core male plants of kiwifruit in this study could provide help for the identification and breeding of kiwifruit varieties, and provide a scientific theoretical basis for the protection of intellectual property rights in the future as well. In this research, we selected 100 pairs of SSR primers from kiwifruit SSR database, and used 4 accessions of genetically distant kiwifruit germplasms to screen. We screened out 14 primers, which with most distinctive bands of amplification, high polymorphism and good repeatability. Then we amplified the 33 core male plants of kiwifruit for test in this research, and tested the PCR products by polyacrylamide gel electrophoresis with 10% concentration. A total of 171 bands were amplified from 14 pairs of SSR primers, of which 163 bands (95.32%) were polymorphic bands. The 33 accessions of kiwifruit germplasms could be completely distinguished by at least 5 efficient pairs of SSR primers. A total of 83 bands were generated from these 5 pairs of SSR primers, of which 82 bands (98.80%) were polymorphic bands. The number of alleles at each locus was between 1 and 12, with an average of 16.6 alleles for each pair of primers. Through the identification of 33 male kiwifruit germplasm resources, excellent marker loci of male kiwifruit germplasm, such as UDK 96-035, UDK 96-053, UDK 96-040, ThrS01 and Forl3, were explored,which could provid firm theoretical basis for future research.
