摘要
研究艳山姜组织培养,为其种苗快速繁育和应用提供技术支持。以种子作为外植体,通过比较种子处理方式,优化培养基的蔗糖浓度和植物激素以及组培苗的移栽基质,建立起艳山姜的组织培养技术。结果表明:破损处理有利于种子的萌发,发芽率可达到93.8%;培养基MS+6-BA 3.0 mg/L+NAA 0.2 mg/L+蔗糖30 g/L对丛芽生长影响显著,培养30 d的增殖系数达到4.93;培养基MS+NAA 0.8 mg/L+蔗糖30 g/L诱导生根最佳,生根率为100%,平均生根5.58条;组培苗移栽于椰糠和椰糠+表土+河沙的基质中,成活率达到100%。该种子组织培养技术适用于艳山姜种苗的规模化生产和应用。
Study on the tissue culture of Alpinia zerumbet was used to provide references for the rapid propagation and application of seedlings. Using the seeds as explants, the technique of tissue culture of Alpinia zerumbet was established by comparing seed treatment, optimizing sucrose concentration and plant hormones of culture medium and transplant substrates of tissue culture seedlings. The results showed that damage treatment was good for seed germination, germination rate was 93.8%. MS +6-BA 3.0 mg/L +NAA 0.2 mg/L +sucrose 30 g/L could significantly influence fasciculate buds, and the propagation coefficient reached 4.93 after 30 days. The most optimum rooting medium was MS+NAA 0.8 mg/L+sucrose 30 g/L, the rooting rate was 100% and the average root number was 5.58. The tissue culture seeding were transplanted in coconut chaff and coconut chaff +topsoil +river sand, and the survival rate were 100%. The seed tissue culture technology might be suitable for the large-scale production and application of Alpinia zerumbet.
