急性胰腺炎合并肺损伤的孕期大鼠组织中Bax及Bcl-2表达的变化及意义分析 被引量:2

Expression and significance of Bax and Bcl-2 in tissues of rats with acute pancreatitis combined with lung injury

摘要 目的:探讨与分析急性胰腺炎(AP)合并肺损伤(LI)的孕期大鼠组织中Bax及Bcl-2表达的变化及意义。方法:Wistar孕期大鼠组12只随机分为对照组、AP组和AP-LI组,每组各4只。大鼠AP-LI模型采用牛磺胆酸钠自胆胰管注入建立,AP组给予甲强龙预防LI。3组在建模后12h进行胰腺、肺损伤病理评分,测定肺组织内细胞凋亡,RT-PCR法测定肺组织Bax及Bcl-2的表达。结果:对照组孕鼠一般情况较好,解剖胰腺和肺外观无明显改变;AP组与AP-LI组一般情况较差,肺表面可见暗红色斑块,解剖胰腺质硬。AP-LI组、AP组的胰腺与肺组织病理评分明显高于对照组,AP-LI组的评分也高于AP组,对比差异都有统计学意义(P<0.05)。对照组Bax及Bcl-2mRNA的表达均很弱,AP-LI组(P<0.05)、AP组的Bax及Bcl-2mRNA表达较对照组各时点均明显增强,P-LI组也明显高于AP组(P<0.05)。AP-LI组、AP组、对照组的细胞凋亡指数分别为(26.33±3.22)%、(7.29±1.44)%和(1.33±0.21)%,3组对比差异均有统计学意义(F=7.392,P<0.05)。结论:AP合并LI可增强孕期大鼠Bax及Bcl-2基因的表达,促进细胞凋亡,肺组织细胞凋亡与Bax、Bcl-2基因表达参与了AP合并LI发病机制。 Objective:To investigate and analysis the significance of Bax and Bcl-2 expression in the tissues of rats with acute pancreatitis(AP)combined with lung injury(LI).Method:Twelve Wistar pregnancy rats were randomly divided into control group,AP group and AP-LI group,4 rats in each group.The AP-LI model of rats were injected with sodium taurocholate into the bile duct,and the AP group was given methylprednisolone to prevent LI.In the three groups,the pathological scores of pancreas and lung injury were determined after construction model of 12 h,and the apoptosis in lung tissue were measured.The mRNA expression of Bax and Bcl-2 in lung tissue was measured by RT-PCR.Result:In the control group,the pregnant mice were in good condition,and the appearance of anatomy pancreas and lung were not changed.The AP group and the AP-LI group were generally poor,and the dark red patches on the lung surface were found,and the pancreas was hard to dissect.The pathological scores of pancreas and lung in AP group and the AP-LI group were obviously higher than those in control group,and the scores of the AP-LI group was higher than that of the AP group,the difference were statistically significant(P〈0.05).The mRNA expression of Bax and Bcl-2 in the control group were very weak,and the mRNA expression of Bax and Bcl-2 in the AP-LI group and AP group were significantly higher than those in the control group(P〈0.05),and the AP-LI group were also significantly higher than that of the AP group(P〈0.05).The apoptotic indexes in the AP-LI group,AP group and control group were(26.33±3.22)%,(7.29±1.44)%and(1.33±0.21)% respectively,and the difference among the three groups were statistically significant(F=7.392,P〈0.05).Conclusion:AP combined with LI can enhance the expression of Bax and Bcl-2 genes and promote apoptosis in rats during pregnancy,and the apoptosis of lung tissue and the expression of Bax and Bcl-2 genes are involved in the pathogenesis of AP combined with LI.
