槲皮素对宫颈癌细胞SiHa迁移和侵袭能力的影响 被引量:3

摘要 目的探讨槲皮素对人宫颈癌SiHa细胞迁移和侵袭能力的影响。方法 SiHa细胞经过不同浓度的槲皮素(0、20、40和80μmol/L)分别处理后,通过Transwell小室检测SiHa细胞迁移和侵袭能力的变化;通过Western blotting检测槲皮素对迁移和侵袭相关蛋白MMP9表达的影响。结果 SiHa细胞经过不同浓度的槲皮素(0、20、40和80μmol/L)分别处理后,可以显著抑制SiHa细胞的迁移能力,每视野下的细胞数分别为202.00±11.66、148.80±14.01、46.00±15.36和13.80±3.96。SiHa细胞经过不同浓度的槲皮素(0、20、40和80μmol/L)分别处理后,可以显著抑制SiHa细胞的侵袭能力,每视野下的细胞数分别为138.00±13.40、99.80±14.72、33.20±8.13和9.80±2.77。SiHa细胞经过槲皮素处理后,可以明显下调MMP9蛋白的表达。结论槲皮素通过下调SiHa细胞中MMP9蛋白的表达,可以抑制SiHa细胞的迁移和侵袭。 Objective To investigate the effect of quercetin on migration and invasion of human cervical cancer cell SiHa. Methods Transwell chamber was used to detect the effect of quercetin on migration and invasion of cervical cancer cell SiHa at different concentrations(0, 20, 40 and 80μmol/L, respectively). Western blotting was used to detect the expression of MMP9 in quercetin treated SiHa cells. Results After treated with different concentrations of quercetin(0, 20, 40 and 80μmol/L, respectively), inhibit migration ability of SiHa cells could be inhibited remarkably, and the cell number was 202.00±11.66,148.80±14.01, 46.00 ± 15.36 and 13.80±3.96, respectively per view. After treated with different concentrations of quercetin(0, 20, 40 and 80μmol/L, respectively), invasion ability of SiHa cells were inhibited significantly, and the cell number was 138.00 ± 13.40, 99.80± 14.72, 33.20± 8. 13 and 9.80 ± 2. 77, respectively per view. Western blotting revealed that the expression of MMP9 protein was significantly down-regulated by SiHa cells treated with quercetin. Conclusion Quercetin could inhibit cell migration and invasion by down-egulating the expression of MMP9 in cervical cancer cell SiHa.
