梨蔗糖合成相关酶SUS和SPS基因家族的鉴定与表达分析 被引量:18

Genome-wide Identification and Expression Analysis of Sucrose Synthase(SUS)and Sucrose Phosphate Synthase(SPS)Gene Families in Pear

摘要 运用生物信息学方法,对梨蔗糖合酶(SUS)和蔗糖磷酸合成酶(SPS)基因进行全基因组鉴定与基因结构、系统发育关系、蛋白质保守功能域、染色体定位和基因进化模式分析,利用转录组数据和实时荧光定量PCR技术分析低蔗糖型‘砀山酥梨’和高蔗糖型‘翠冠’果实发育过程中SUS和SPS家族基因的表达模式。结果表明,梨中包含17个SUS基因(Pbr SUS1~Pbr SUS17)和8个SPS基因(Pbr SPS1~Pbr SPS8);SUS基因家族分为3个亚家族,分别为SUS1、SUSA和SUS2。梨SUS和SPS基因家族成员不均匀地分布在10条染色体上,主要通过片段复制事件进行扩张,且在进化过程中主要受纯化选择作用。Pbr SPS3、Pbr SPS4、Pbr SPS8、Pbr SUS2和Pbr SUS15是调控梨果实蔗糖合成积累的重要基因。 In this study,the sucrose synthase(SUS)and sucrose phosphate synthase(SPS)gene families were identified from pear(Pyrus bretschneideri‘Dangshan Suli') genome database,and their gene structure,phylogenetic relationship,protein conserved domains,chromosomal locations and gene evolution pattern were analyzed by bioinformatic approaches. Transcriptome data and quantitative real-time PCR were used separately to analyze the expression patterns of SUS and SPS gene families during the fruit development of low-sucrose‘Dangshan Suli'and high-sucrose‘Cuiguan'(P. pyrifolia). The results showed 17 SUS genes(Pbr SUS1–Pbr SUS17)and 8 SPS genes(Pbr SPS1–Pbr SPS8)were identified in P. bretschneideri,including 3 subfamilies for SUS gene family(SUS1,SUSA and SUS2). The expansion of Pbr SUS and Pbr SPS gene families distributed unevenly on 10 chromosomes were mainly caused by segmental duplication,and their evolutionary processes were affected by purify selection. Pbr SPS3,Pbr SPS4,Pbr SPS8,Pbr SUS2,and Pbr SUS15 are the important genes in regulating the synthesis and accumulation of sucrose for pear fruit.
