多成分含量测定结合UPLC指纹图谱及其模式判别评价子芩与枯芩质量 被引量:9

摘要 目的:建立子芩与枯芩的UPLC指纹图谱及其分析方法,从整体成分分析子芩与枯芩饮片质量的专属性差异。方法:采用UPLC法测定不同产地的子芩与枯芩样品,对比了黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、千层纸素A 5种成分的含量变化;分析两者UPLC的专属性差异特征和相似度评价,并采用主成分分析和偏最小二乘法判别20批子芩与枯芩的指纹图谱。结果:子芩中黄芩苷和汉黄芩苷含量较枯芩分别高28.87%、54.12%,其中子芩的黄芩苷含量范围在11.60%~13.03%之间,枯芩在8.01%~10.53%之间;子芩中黄芩素、汉黄芩素、千层纸素A含量较枯芩明显较低,分别低70.37%、44.44%、59.09%;通过主成分分析、偏最小二乘判别分析发现子芩与枯芩差异明显,且不同产地也存在一定差异。结论:子芩和枯芩中黄芩苷、汉黄芩苷、黄芩素、汉黄芩素以及千层纸素A的含量具有明显差异,其UPLC指纹图谱中黄芩苷(3号峰)与黄芩素(10号峰)峰面积比具有明显差异,具鉴别意义,同时其化学计量模式的识别也为子芩和枯芩的质量评价及品质研究提供了一定的参考。 Objective: To established UPLC fingerprints and analysis method,to analyze the difference of the specificity between Scutellaria baicalensis Pith-nodecayed and Scutellaria baicalensis Pith-decayed from the whole compositions. Methods: The samples of the Scutellaria baicalensis Pith-nodecayed and Scutellaria baicalensis Pith-decayed in different producing areas were determined by UPLC,the contents of five components as baicalin,wogonoside,baicalein,wogonin,oroxylin A were compared; By using the PCA and PLSDA method,the fingerprints of 20 batches of Scutellaria baicalensis Pith-nodecayed and Scutellaria baicalensis Pith-decayed were identified by analyzing the difference of the specificity and similarity evaluation of UPLC. Results: The contents of baicalin and wogonoside in Scutellaria baicalensis Pith-nodecayed were higher than that in Scutellaria baicalensis Pith-decayed as 28. 87%,54. 12% respectively,the contents of baicalin in Scutellaria baicalensis Pith-nodecayed were range from 11. 60% - 13. 03%,while the Scutellaria baicalensis Pith-decayed was between 8. 01% - 10. 53%; However,the contents of baicalein,wogonin,oroxylin A were significantly lower as70. 37%,44. 44%,59. 09% respectively. There were significant difference through the PCA and PLS-DA. Conclusion: The contents of baicalin,wogonoside,baicalein,wogonin,oroxylin A were siginificantly different between Scutellaria baicalensis Pith-nodecayed and Scutellaria baicalensis Pith-decayed; and the radio of baicalin( 3) and baicalein( 10) peak area has obviously differences. Meanwhile,the identification provides somereference for the quality evaluation and quality research.
