Optimization of Infrared Assisted Fast Extraction of Flavonoids from Scutellaria baicalensis by BBD-RSM Combined with Multi-index Approach

摘要 目的:以多指标法筛选黄芩黄酮的最佳红外工艺参数。方法:采用UHPLC法建立黄芩黄酮的含量测定方法;基于单因素结果,结合三因素三水平Box-Behnken Design(BBD)-Response Surface Methodology(RSM)试验设计,比较分析3种指标(综合评分值、总评归一值和总黄酮含量值)与自变量之间的数学关系,经响应面法预测最佳提取条件。结果:综合评分法与总黄酮含量法的分析结果一致,且在模型中其各项指标均优于总评归一值法。最佳工艺为:乙醇浓度60%,提取时间6.25 min,料液比1∶75,提取功率200 W。最优工艺条件下的试验测定值与模型预测值相吻合。结论:采用BBD-RSM法优选黄芩黄酮的红外提取工艺,操作简便,预测性能好,可为黄芩黄酮提取研究提供一条新途径。 Objective: To screen the optimum parameters of infrared extraction of flavonoids from Scutellaria baicalensis by multiindex approach. Methods: Determination of flavonoids was established by UHPLC method. Based on the single-factor experiment,a three factor and three level Box-Behnken Design( BBD)-Response Methodology Surface( RSM) experimental design was used to compare mathematical relationship between three different indicators( comprehensive score,overall desirability value method and total flavonoid content) and the variables,and to forecast the optimum extraction condition. Results: The result obtained by the method of total flavonoid content was similar with the comprehensive score method,and the model parameters were better than that of the overall desirability method. The optimum parameters was as follows,ethanol concentration was 60%,the extraction time was 6. 25 min,material to liquid ratio was 1∶75,and extraction power was 200 W. Under the optimum condition,the experimental results were consistent with the predicted values. Conclusion: The best extraction process of flavonoids from Scutellaria baicalensis by BBD-RSM method has simple operation and good predict performance,which can provide a new way for the extraction process of flavonoids from Scutellaria baicalensis.
