摘要
目的:探讨肾气丸对衰老大鼠小肠干细胞及其微环境Wnt/β-catenin信号通路的调节作用。方法:使用SD大鼠,根据不同发育阶段分为6月龄成年对照组、22月龄衰老模型组及肾气丸低、高剂量组。采用HE染色观察小肠基本形态及测量相应指标,采用免疫组织化学法检测各组大鼠小肠PCNA、Bmi 1、β-catenin、GSK-3β的表达情况。结果:与成年组比较,衰老模型组绒毛长度、绒毛宽度、隐窝深度、绒毛/隐窝值(V/C值)均显著降低,PCNA、Bmi 1、β-catenin表达显著降低,而GSK-3β表达显著升高。与衰老模型组比较,肾气丸各组绒毛长度、绒毛宽度、隐窝深度、绒毛/隐窝值(V/C值)均显著升高,PCNA、Bmi 1、β-catenin表达显著均升高,GSK-3β表达显著降低。结论:肾气丸可改善衰老大鼠小肠干细胞增殖功能并上调Wnt/β-catenin信号通路相关蛋白表达。
Objective: To explore the effect of Shenqi pills on small intestinal stem cells and the microenvironment of Wnt/β-catenin signaling pathway in aging rats. Methods: SD rats were divided into six month age as adult control group,22 month age as aging model group,Shenqi pill low-dose group,Shenqi pill high-dose group. The morphology of small intestine was determined by HE staining and the corresponding indexes were measured. The expression of PCNA,Bmi 1,β-catenin and GSK-3β in small intestine was detected by immunohistochemistry. Results: Compared with the adult control group,the villus length,villus width,crypt depth,and villus/crypt value( V/C) in aging group were decreased significantly. The expression levels of PCNA,Bmi 1 and β-catenin were decreased significantly,while the expression of GSK-3β was increased significantly in the intestine of the aging group. Compared with aging group,the villus length,villus width,crypt depth and villus/crypt value( V/C) of shenqi pill groups were increased significantly. The expression levels of PCNA,Bmi 1 and β-catenin were increased significantly,while the expression of GSK-3β was decreased significantly in the intestine of the aging group. Conclusion: Shenqi Pills can improve the proliferation of intestinal stem cells and upregulate Wnt/β-catenin signaling pathway in aging rats.
