摘要
目的 研究白血病NB4细胞对白蛋白纳米粒的摄取行为,探讨其靶向给药白血病细胞的可行性。方法 以香豆素-6示踪白蛋白纳米粒,采用激光共聚焦显微镜与流式细胞术法考察人急性早幼粒白血病NB4细胞对纳米粒的摄取行为,进而以MTT比色法评价载二氢丹参酮Ⅰ白蛋白纳米粒对细胞的增殖抑制作用。结果 与游离药物香豆素-6、二氢丹参酮Ⅰ相比,白蛋白纳米粒可显著增加药物细胞对药物的摄取,增强其对NB4细胞的增殖抑制作用。结论 白蛋白纳米粒对NB4细胞具有靶向性,有望成为白血病靶向给药的新型载体。
OBJECTIVE To investigate the uptake of albumin nanoparticles (NPs) in acute promyelocytic leukemiacell NB4 and explore the possibility of NPs as a novel drug delivery vehicle to target the leukemia cells. METHODS The bovine serum albumin nanoparticles( BSA- NPs)were prepared and labelled by a fluorescent marker, coumarin -6, and then its uptake by NB4 cells was analyzed with laser confocal microscope and flow cytometry. Dihydrotanshinone I loaded BSA - NPs was developed and its anti - tumor effect on Ntl4 cells was evaluated by MTI" assay. RESULTS Compared with free coumarin - 6 or dihydrotanshinone I , BSA - NPs significantly increased the cellular uptake of encapsulated drug and enhanced the inhibitory effect on the proliferation NtM cells. CONCLUSION The albumin NPs were took by NB4 ceils were confirmed,thus delivering more drugs into the cells. It suggests that the albumin NPs may serve as a promising drug delivery vehicle that targeting leukemia cells.
