交通心肾中药对更年期大鼠性激素及神经递质的影响 被引量:14

Effects of Jiaotong Xinshen Decoction of coordinating heart and kidney on sex hormone and neurotransmitters in climacteric rats

摘要 目的:探讨交通心肾中药对更年期模型大鼠去甲肾上腺素(NA)、5-羟色胺(5-HT)、β-内啡肽(β-EP)等神经递质以及雌二醇(E_2)、促卵泡刺激素(FSH)、促黄体生成素(LH)等性激素的影响。方法:选用12-13月龄雌性SD大鼠及行卵巢去势手术的3月龄雌性SD大鼠作为更年期模型,随机分为老龄空白组、老龄中药组、老龄西药组、去势空白组、去势中药组、去势西药组,分别给予0.9%氯化钠溶液、交通心肾方药(15g/kg)、戊酸雌二醇(0.1mg/kg)灌胃,共28d,另选3月龄雌性SD大鼠作为青年鼠组予0.9%氯化钠溶液。用药结束后,采用ELISA法检测各组大鼠血清E_2、FSH、LH、NA、5-HT、β-EP水平。结果:交通心肾中药"宁更丹"和戊酸雌二醇对两种模型大鼠均可显著升高血清E_2,降低FSH和LH(P<0.05),同时有升高NA和β-EP水平、降低5-HT水平的趋势。结论:去势手术更年期模型更能反映应激对绝经综合征的影响;交通心肾中药可调节更年期大鼠中枢神经系统和内分泌网络功能,可能是其治疗女性绝经综合征的作用机制之一。 Objective: To explore the influence of therapy of coordinating heart and kidney on norepinephrine(NA), 5-hydroxytryptamine(5-HT) and beta endorphin( 13 -EP) and estradiol(E2), follicle stimulating hormine(FSH), luteinizing hormone(LH), etc. in climacteric rats. Methods: 12-13 months old female SD rats were randomly divided into aging group, TCM aging group, western drug aging group, and meanwhile, ovariectomized 3 months old female SD rats as menopause models were randomly divided into castrated group, TCM castrated group and western drug castrated group. Aging and castrated group were separately given saline, TCM aging and Jiaotong Xinshen Decoction (15g/kg), Estradiol Valerate (0.1mg/kg) by gavage for 28 days. Other 3 months old female SD rats were chosen as young group given with saline. The levels of E2, FSH, LH, NA, 5-HT and β -EP in serum were detected by ELISA. Results: Both of the Jiaotong Xinsben Decoction of 'Ning Gengdan' and Acid Estradiol could significantly elevate the levels of E2, and decreased the levels of FSH, LH in serum (P〈0.05), and there were the trend of increasing NA and β -EP and decreasing 5-HT. Conclusion: The surgical castration of menopausal model can reflect the influence of stress on menopausal syndrome better than the aging model; the therapy of coordinating heart and kidney can regulate the central nervous system and endocrine the network function in climacteric rats, which may be one of the mechanisms in the treatment of female menopausal syndrome.
