新扶正除疫颗粒对肺炎链球菌所致上呼吸道感染小鼠脾组织TLR2、TLR4 mRNA的影响 被引量:3

Effects of New Fuzheng Chuyi Granules on spleen tissue TLR2 and TLR4 mRNA of mice with a cute upper respiratory infection caused by streptococcus pneumonia

摘要 目的:以肺炎链球菌鼻腔滴入导致急性上呼吸道感染的小鼠为模型,采用Real-time PCR方法,检测脾脏组织中Toll样受体(TLR)2、TLR4 m RNA的变化,探讨新扶正除疫颗粒中药复方在抗炎及免疫调节方面作用机制。方法:BALB/c小鼠随机分为空白组,模型组,复方高、中、低剂量组,中药对照组和西药对照组,造模1h后灌药,第1、4、7天为取材时间点,测定小鼠脾组织中脾脏指数及TLR2、TLR4 m RNA表达情况。结果:药物干预后各时间点脾脏指数均有不同程度下降。模型组脾脏TLR2、TLR4 m RNA表达水平相对空白组明显增强(P<0.05),且随感染时间的增加表达量逐渐增强。复方低、中、高剂量组表达水平逐渐下降,复方高剂量组表达水平随感染时间下降趋势最明显。结论:新扶正除疫颗粒可通过调节脾脏TLR2、TLR4 m RNA表达,调控机体抗感染免疫作用,对激活第一道天然免疫的屏障具有一定作用。 Objective: By taking the mice with acute upper respiratory infection caused by streptococcus pneumonia dripped from nasal cavity as modal, and adopting Real-time PCR method, and detecting the change of TLR2 and TLR4 mRNA in spleen tissue to discuss the mechanism of New Fuzheng Chuyi Granules in anti-inflammation and immunoregulation. Methods: BALB/c mice were devided into blank group, model group, New Fuzheng Chuyi Granules high and medium and low dose group, Traditional Chinese Medicine control group and Western Medicine control group randomly, and were given medicine lh after molding. Tissues were taken at ld, 4d, 7d, measuring TLR2 and TLR4 mRNA in mice spleen tissue. Results: There was a significant increase of expression level of spleen TLR2 and TLR4 mRNA in model group compared with blank group (P〈0.05), and there was a increasing tendency in expression level with infection time going on. There was a decrease of expression level in New Fuzheng Chuyi Granules high and medium and low dose group, and the most significant decreasing tendency in expression level came with the high dose group with infection time going on. Conclusion: New Fuzheng Chuyi Granules contributes to adjusting and controlling body anti-infectious immunity, and activating the first barrier of body's immune system through regulating spleen TLR2 and TLR4 mRNA.
