基于普通病理学及abcc1基因研究全天麻胶囊及天麻素对卡马西平抗癫痫作用的影响 被引量:2

Study on the effects of the Gastrodia Capsule and the gastrodin combined Carbamazepine to PG-kindled rat by general pathology and abcc1 gene

摘要 目的:研究全天麻胶囊及天麻素联合卡马西平后,对卡马西平抗癫痫药效学的影响,同时研究多次给予全天麻胶囊、天麻素、卡马西平后,对癫痫大鼠大脑脑部缺血引起的神经元损伤及abcc1表达水平的影响。方法:采用青霉素点燃的癫痫大鼠模型,通过癫痫评分评价全天麻胶囊及天麻素联合卡马西平抗癫痫作用,通过普通病理学、RT-PCR分别在给予对应药物第1、7、15天,对各组癫痫大鼠脑部海马体神经元及abcc1表达量进行测定。结果:全天麻胶囊及天麻素可提高卡马西平的抗癫痫作用。各组神经元细胞缺血性改变、核固缩、核仁溶解、胞浆空泡等随时间改善,研究发现给予全天麻胶囊改善效果明显优于其它未给予组,但天麻素及卡马西平则无该类作用。各组大鼠大脑海马体abcc1基因表达随之增加。结论:全天麻胶囊可通过影响癫痫大鼠脑部神经元及abcc1基因的表达来增强卡马西平抗癫痫作用,但是否有其它作用机制尚不明确。 Objective: This study was designed to illustrate the antiepileptic effect and possible mechanism of Gastrodia Capsule and Gastrodin combined with Carbamazepine on Penicillins (PG)-kindled rat, and further revealed the influence of Gastrodia Capsule and gastrodin on the antiepileptic action of Carbamazepine. Addti0nally, the article also studied the effect of multiple dosage of Gastrodia Capsule, gastrodin and Carbamazepine on the neurons injury and the expression level of abccl on epileptic rat brain. Methods: This study Used PG-kindled rat as epileptic model and scored the state of epileptic seizure of PG- induced epilepsy rat, to measure the antiepileptic effect of Gastrodia Capsule and gastrodin combined with Carbamazepine. Then we used general pathology and RT-PCR methods to study the effect of Gastrodia Capsule and gastrodin combined with Carbamazepine on the hippocampus nerve cells and the expression level of abccl at first day,7th day,15th day. Results: The Gastrodia Capsule and gastrodin could significantly increased the antiepileptic effect of Carbamazepine. The results found that Gastrodia Capsule could significantly improve the status of neurons with ischemic changes, nucleus pycnosis, nucleolus dissolved and cytoplasm vacuoles with the passage of time. But gastrodin and Carbamazepine couldn't. Further, the results showed that the expression level of abcel in all group were all up-regulated by treatment. Conclusion: The Gastrodia Capsule can significantly increased the antiepileptic effect of Carbamazepine, and can attenuate the damage of neurons and increase the expression level of abccl. Nonetheless, the underlying mechanism for antiepileptic activity of Gastrodia Capsule remains to be further investigated.
