摘要
目的:观察加减三甲散对肝纤维化大鼠肝脏Th17/Treg平衡的调控作用,从免疫学的角度探索其作用机制。方法:选取健康雄性大鼠18只,随机分成正常组、模型组、治疗组(加减三甲散组)。采用猪血清腹腔注射法复制大鼠肝纤维化模型,治疗组以加减三甲散干预,治疗60 d后检测各组大鼠肝纤维化指标、Th17细胞和Treg细胞的相关指标,了解Th17/Treg平衡的变化情况。结果:(1)与正常组比较,模型组大鼠肝纤维化指标LN、Ⅳ-C、PCⅢ、HA明显升高(P〈0.05)。(2)模型组大鼠RORγt、Foxp3、IL-6,IL-17、IL-10、RORγt/Foxp3、IL-6/IL-10、IL-17/IL-10均较正常组显著升高(P〈0.01或P〈0.05),加减三甲散组上述指标均较模型组显著降低(P〈0.01或P〈0.05)。结论:加减三甲散可减轻大鼠肝纤维化程度,其可能机制是通过下调肝组织中Th17及Treg细胞数量,使Th17/Treg平衡偏于Treg方向发展,从而有利于肝纤维化的控制及逆转。
Objective: To investigate the influence of Modified Sanjia Powder on the balance of Th17/Treg in the liver of rats with hepatic fibrosis and explore its mechanism from the perspective of immunology. Methods: Eighteen healthy male rats were divided randomly into normal group,model groupand and treatment group( Modified Sanjia Powder group). Rat liver fibrosis model was induced by intraperitoneal injection of porcine serum. The treatment group was treated with Modified Sanjia Powder. After 60 days of treatment,the indexes of hepatic fibrosis and related indexes of Th17 and Treg cell were detected to learn the change of Th17/Treg balance. Results:( 1) Compared with normal group,the fibrosis index LN,Ⅳ-C,PC Ⅲ and HA of model group were significantly higher( P 〈0. 01). Compared with the model group,the fibrosis index LN,PCⅢ and HA of Modified Sanjia Powder group decreased( P 〈0. 01 or P 〈0. 05) and there was no significant in Ⅳ-C( P 〈0. 05).( 2) Compared with normal group,RORγt,Foxp3,IL-6,IL-17,IL-10,RORγt/Foxp3,IL-6/IL-10 and IL-17/IL-10 in the liver tissue of rats in the model group increased( P 〈0. 01 or P 〈0. 05). Compared with model group,all the indexes in the Modified Sanjia Powder group decreased( P 〈0. 01 or P 0. 05). Conclusions: Modified Sanjia Powder could reverse the process of liver hepatic fibrosis in rats to a certain extent. Its possible mechanism is by down regulating the number of Th17 and Treg cell in liver tissue of rats and making the balance of Th17/Treg in the direction of Treg,which was beneficial to the control and reversion of liver fibrosis.
