摘要
利用白菜SSR引物对56份菜心种质资源进行遗传多样性分析。结果表明:19对SSR引物扩增出131条等位基因,平均每对引物扩增出6.89个位点,说明白菜SSR引物在菜心遗传分析中具有通用性;多态性位点百分率P、基因杂合度(He)、有效等位基因数(Ne)、Shannon.Wiener指数(H′)和多态信息含量(PIC)的均值结果分别为:93.03%、0.884、10.344、2.340、0.870,各指标间相关性极显著;聚类结果与遗传相似性的相关性r=0.746,说明聚类图可以大致描述菜心资源分类;56份菜心聚为2大类:第1类多为地方特色菜心,属于白菜薹类型,第Ⅱ类群为菜心商品种类型,深色菜心是浅叶材料的进化方向,但要注意系谱选育的自交代数。
The genetic diversity of 56 Chinese flowering cabbage genetic resources was analyzed by using cabbage SSR primer. The results showed thatl9 cabbage SSR primer combinations amplified 131 alleles and each pair of primers amplified 6.89 loci on average, which indicated that cabbage SSR primers were versatile in genetic diversity analysisof Chinese flowering cabbages. The polymorphism loci percentage (P), genetic heterozygosity (He), the number of effective alleles (Ne), Shannon-Wiener index (H′) and polymorphism information content (PIC) were 93.03%, 0.884, 10.344, 2.340 and 0.870, respectively.The correlation among the indexes was extremely significant. The correlation between clustering result and genetic similarity was r=0.746, which showed that classification of Chinese flowering cabbage resources could be generally describted by clustering figure.56 flowering Chinese cabbage resources were divided into 2 groups. Cluster ⅠI mostly wasthe local Chinese flowering cabbage which belonged to Cabbage Sprouts type. Cluster Ⅱ was commercialChinese flowering cabbagetype. The color of leaf evolved from light to dark, but the numbers of self-generation based on Pedigree breeding must be paid attention to.
