蝴蝶兰腋芽增殖过程中的转录组特性 被引量:3

Transcriptome Characteristics of Phalaenopsis in Axillary Bud Proliferation

摘要 蝴蝶兰在种苗生产中主要是以腋芽再生的方式进行繁育。本研究以大辣椒、四季春2个蝴蝶兰品种为试材,利用Illumina HiSeq-(XM)2000平台对他们在组织培养过程中3个诱导增殖阶段的转录组进行了测序。结果表明,测序共获得52.77Gb的Clean Data,6个样品的Clean Data平均达到了6.78Gb。对CleanData进行过滤并组装,共获得86130条Unigenes;其中长度在1kb以上的Unigenes有16723条。参照无参基因组在数据库中进行比对分析,其中28430条Unigenes可进行有效注释。对2个品种、3个诱导增殖阶段的全部Unigenes进行SNP分析表明,四季春的杂合性SNP分布密度明显低于大辣椒品种。另外,对转录组数据进行了差异表达基因分析,共获得了7638条差显基因。 The seedlings production of Phalaenopsis is mainly induced and regenerated from axillary bud. In this research, P. Big Chili and P. Spring (Phalaenopsis spp.) were selected as test materials. RNA-sequencing oftranscriptomes at three induced proliferation stages was carried out by using Illumina HiSeq-(TM) 2000 during the process of tissue culture. The results showed that a total of 52.77 Gb Clean Data were obtained, and 6 samples reached an average of 6.78 Gb. A total of 86 130 Unigenes were obtained after Clean Data were filtered and assembled, of which 16 723 Unigenes were over 1 kb in length. Functional annotation scanning in the transcript database showed that a total of 28 430 Unigenes could be effectively annotated. The SNP analysis of two cultivars and the total Unigenes at three induced proliferation stages showed that SNP distribution density of P. Spring heterozygosity was obviously lower than Big Chili. Besides, differentially expressed genes were analyzed and 7 618 differentially expressed genes were obtained.
