氧化苦参碱对小鼠滤泡辅助性T细胞免疫功能的影响 被引量:11

摘要 目的初步探讨氧化苦参碱对小鼠滤泡辅助性T细胞(Tfh)免疫功能的影响。方法 BALB/C小鼠腹腔注射不同剂量的氧化苦参碱,7d后收集脾细胞并分离获得单个核细胞,采用流式细胞术检测Tfh细胞比例;RT-q-PCR方法测定Tfh细胞B细胞淋巴瘤分子6(Bcl-6)的mRNA转录水平;采集小鼠眼球血,ELISA法检测外周血IL-21的表达水平。结果氧化苦参碱的高[180mg·(kg·d)^(-1)]、中剂量组[90mg·(kg·d)^(-1)]、低剂量组[45mg·(kg·d)^(-1)]均可减少BALB/C小鼠脾Tfh细胞的比例(P<0.05),且高剂量组作用强于中、低剂量组(P<0.05);氧化苦参碱的高、中剂量组可抑制Tfh细胞Bcl-6 mRNA的转录(P<0.05),降低小鼠IL-21的分泌水平(P<0.05),且高剂量组的作用强于中剂量组(P<0.05)。结论氧化苦参碱可能具有调节Tfh细胞免疫功能的作用。 Objective To investigate the effect of oxymatrine(OMT)on follicular helper T cells. Methods BALB/C mice were treated with different dose of oxymatrine by intraperitoneal injection for 7 days. The ratio of Tfh cells collected from spleen monocytes was detected by flow cytometry. The level of Bcl-6 mRNA was measured by RT-q-PCR. The expression of IL-21 in peripheral blood was tested by ELISA. Results Compared with N.S group, high dose group [ 180 mg·(kg·d)^-1 ], middle dose group [ 90 mg·(kg·d)^-1 ] and low dose group [ 45 (kg·d)^-1] of OMT decreased ratio of Tfh cells. The high dose group showed more obvious inhibitory effect than the other two dose groups of OMT. High dose group and middle dose group of OMT inhibited the level of Bcl-6 mRNA(P〈0.05) in Tfh cells and the serum expression of IL-21 in the BALB/C mice(P〈0.05). The inhibitory of high dose group was more significant than middle dose group(P〈0.05). Conclusion OMT may regulate immunological function of Tfh cells.
