血浆miR-132和MMP-3的表达及其与类风湿关节炎的关联性研究 被引量:3

摘要 目的检测类风湿关节炎(RA)患者和健康对照组血浆中miR-132和MMP-3的表达,探讨miR-132和MMP-3与类风湿关节炎关联性,为类风湿关节炎诊断提供一定理论依据。方法收集RA患者80例,健康对照64例,Real-Time-PCR检测血浆中miR-132的表达水平,ELISA检测MMP-3的表达水平,并进行相关性分析。结果 RA组患者血浆miR132的表达低于健康对照组,MMP-3的表达高于健康对照组(P均<0.05);不同RA活动度之间miR-132、MMP-3的表达水平差异有统计学意义(P<0.05)。相关分析显示,RA患者血浆中miR-132与MMP-3的表达水平呈负相关性(r=-0.756,P<0.01),对照组血浆中miR-132与MMP-3的表达水平呈负相关性(r=-0.683,P<0.01)。ROC曲线分析显示血浆miR-132的曲线下面积为0.960。结论 RA患者血浆中miR-132的表达降低,MMP-3的表达升高,两者呈负相关,血浆miR-132水平可能成为RA的诊断标志物。 Objective To explore the miR-132 and MMP-3's correlation with RA by detecting the expression ofmiR-132 and MMP-3 in the peripheral blood of patients with rheumatoid arthritis( RA). Methods By collecting 80 cases of RA patients and 64 cases of healthy controls to detect the expression level of miR-132 by Real-Time PCR, the expression level of MMP-3 by ELISA and correlation analysis. Results The expression of miR-132 in patients in group RA was significantly lower than the control group.MMP-3 expression was significantly higher than the control group(P〈0.05). There are difference between the different activities of the expression levels of miR-132 and MMP-3(P〈0.05).Correlation analysis showed that, miR-132 and MMP-3 showed a significant negative correlation(r=-0.756,-0.683,P〈0.01). ROC curve analysis showed that the area under the curve of plasma miR-132 was 0.960. Conclusion miR-132 is low expression in peripheral blood of RA group, and MMP-3 is high expression, the expression level is associated with disease activity, and showed a negative correlation between the two.ROC curve analysis showed that expression of miR-132 levels in peripheral blood can be used as a potential diagnostic index of RA.
