复方三金胶囊对人乳腺癌荷瘤裸鼠瘤组织中线粒体凋亡通路蛋白表达的影响

Effect of Compound Sanjin Capsule on Protein Expressions of Mitochondrial Apoptosis Pathway in Tumor Tissue of Nude Mice Bearing Breast Cancer

目的:通过研究复方三金胶囊对乳腺癌荷瘤裸鼠瘤组织中Caspase-3、Caspase-9、Bax和Bcl-2蛋白表达的影响,探讨复方三金胶囊促肿瘤细胞凋亡的作用,深入研究其抗肿瘤作用机制。方法:将30只裸鼠接种人乳腺癌MCF-7细胞,待瘤体积超过5 mm时,将30只裸鼠随机分为模型组、复方三金胶囊高、中、低剂量组及环磷酰胺对照组,每组6只,连续给药两周后,取瘤组织进行各项指标测定。指标测定选用免疫组化法,分别从瘤组织染色情况和平均光密度值进行数据分析。结果:与模型组比较,复方三金胶囊各组瘤组织中Caspase-3、Caspase-9、Bax阳性细胞数均显著增加;免疫组化测各组蛋白的平均光密度值结果显示,与模型组比较,复方三金胶囊高剂量组中Caspase-9、Bcl-2的表达呈极显著差异(P<0.01),Caspase-3、Bax的表达呈显著差异(P<0.05);复方三金胶囊中剂量组中Bax的表达呈极显著差异(P<0.01),Caspase-9、Bcl-2的表达呈显著差异(P<0.05);复方三金胶囊低剂量组中Caspase-9的表达呈极显著差异(P<0.01)。结论:复方三金胶囊能够显著抑制乳腺癌细胞的增殖作用与促细胞凋亡作用相关。

Objective: To study the effect of compound Sanjin capsule on the protein expressions of Caspase-3,Caspase-9,Bax and BCl-2 in tumor tissue of nude mice bearing breast cancer,and to explore its effect on apoptosis of tumor cell and the mechanism of its role in anti-tumor,thus to find out the target of the medicine. Methods: MCF-7 cell was used to set up breast cancer in 30 nude mice,the mice were randomly divided into the model group,compound Sanjin groups( high-dose,medium-dose,low-dose) and Cyclophosphamide group after the tumor exceeded 5 mm. After two weeks of continuous administration,the tumor tissue was measured. The immunohistochemical method was used to analyze the data of the tumor tissue and the average optical density. Results: Positive cells of Caspase-3,Caspase-9 and Bax were significantly increased in the compound Sanjin groups,compared to the model group. The average optical density of the histones showed that there were significant differences between the compound Sanjin group of high-dose and the model group in Caspase-9 and Bcl-2( P < 0. 01),as well as in Caspase-3 and Bax( P < 0. 05).There were significant differences between the compound Sanjin group of medium-dose and the model group in Bax( P < 0. 01),as well as in Caspase-9 and Bcl-2( P < 0. 05). There were significant differences between the compound Sanjin group with low-dose and the model group in Caspase-9( P < 0. 01). Conclusion: Compound Sanjin capsule can significantly inhibit the cells proliferation of breast cancer and promote the apoptosis.
