不同配伍补阴方对衰老大鼠肾、肠组织抗氧化能力及P_(16)、P_(21)蛋白表达影响的比较研究

Comparative Study of Tonifying Yin Formulas with Different Compatibility in Aging Rats:Antioxidant Capacity and Protein Expression of P_(16) and P_(21) in Kidney and Intestines

目的:观察左归丸和六味地黄丸及"三补"对D-半乳糖(D-gal)致亚急性衰老大鼠的抗自由基能力及P_(16)、P_(21)蛋白表达的影响,比较两方的不同配伍方式抗衰老机制的差异;通过对小肠组织衰老相关指标变化的观察,初步探讨补肾滋阴方滋腻之性可能会产生的影响。方法 :观察左归丸、六味地黄丸及两方共有"三补"药物组对D-半乳糖(D-gal)致亚急性衰老大鼠血清、小肠和肾组织的总抗氧化能力(T-AOC)、过氧化氢酶(CAT)含量及对小肠、肾组织P_(21)、P_(16)蛋白表达的影响。结果:亚急性衰老大鼠总抗氧化能力(T-AOC)和过氧化氢酶(CAT)水平下降,P_(21)和P_(16)蛋白表达水平上调;三个中药组均能提高血清的T-AOC,六味地黄丸还能提高肠组织的T-AOC,左归丸和和"三补"药物组对小肠和肾组织的T-AOC无提高作用;六味地黄丸和"三补"药物组可以升高血清的CAT的活性,但对小肠和肾组织的CAT无明显作用,左归丸对CAT无明显升高作用;三个中药组均能下调衰老相关基因P_(16)和P_(21)在小肠和肾脏组织上的蛋白表达,并且对于小肠组织的P_(16)和P_(21)蛋白表达的下调作用更为明显。结论:左归丸和六味地黄丸尽管存在补泻配伍方式的不同,但在抗衰作用上差异尚不显著;补泻兼顾和纯补无泻的组方思想只是在具体的药理机制上有所不同,增加观察指标和观察角度是否会出现较为明显的区别值得我们去进一步探索;补益药滋腻碍胃之性可能对小肠的衰老相关指标的变化产生了一定的影响。

Objective: To observe of the effect of Zuogui pill,Liuwei Dihuang pill and Sanbu group,which is the main part of the two formulas,on anti-radical activities and the protein expressions of P_(21) and P_(16) in sub-acute aging rats induced by D-galactose; to compare the different anti-aging mechanism of the formulas with different compatibility;and to explore the effect of greasy property caused by tonifying kidney-yin by observing the changes of relative aging indexes in small intestine. Methods: Observing the effect of Zuogui pill,Liuwei Dihuang pill and Sanbu group on total antioxidant capacity( T-AOC),content of catalase( CAT),and protein expressions of P_(21) and P_(16) in sub-acute aging rats induced by D-galactose. Conclusion: T-AOC and CAT were cut down and the protein expressions of P_(21) and P_(16) were up-regulated in sub-acute aging rats. The serum T-AOC was increased in the three medication groups; Liuwei Dihuang pill could also improve T-AOC of small intestines; however,Zuogui pill and Sanbu group had no effect on T-AOC of small intestine and kidney tissue. Liuwei Dihuang pill and Sanbu group could increase the activity of serum CAT,but had no obvious effect on CAT of small intestine and kidney tissue,and Zuogui pill had no obvious effect on CAT. Protein expressions of P_(16) and P_(21) in small intestine and kidney were down-regulated in all the three medication groups,and the down-regulation was more obviously in small intestine. Conclusion: Although there was compatibility difference between Zuogui pill and Liuwei Dihuang pill,less difference has been observed in anti-aging. The thought of the combination of reinforcing and reducing,or pure supplement only shows difference in the specific pharmacological mechanism. It is worth further exploration to see whether there will be a significant difference with more observing indexes and from different observing angles. Tonics influencing the action of stomach may have some influence on the changes of small intestine related indicators in aging rats.
