摘要
为研究爪哇黄芩二氢黄酮活性成分的抗肿瘤作用,筛选了爪哇黄芩活性部位和敏感细胞株,并研究了爪哇黄芩活性二氢黄酮对敏感细胞株HepG-2细胞增殖的影响,为开发天然药物奠定了实验基础.选取体外培养的人肝癌细胞HepG-2、胃癌细胞SGC-7901、肺癌细胞A549、乳腺癌细胞MCF-7及神经胶质瘤细胞C6为模型,以半数抑制浓度(IC50)为指标,采用MTT法测定爪哇黄芩石油醚、二氯甲烷、正丁醇提取部位对5种肿瘤细胞的增殖抑制作用,结果表明,爪哇黄芩二氯甲烷部位为其活性部位,人肝癌细胞HepG-2为其敏感细胞株.然后,采用MTT法测定活性部位分离的二氢黄酮单体对HepG-2的抑制作用,进行活性单体的筛选.结果显示,爪哇黄芩5,6,2'-三羟基-7-甲氧基二氢黄酮IC_(50)为110.3μmol/L,爪哇黄芩5-羟基-6,7,2',6'-四甲氧基二氢黄酮IC_(50)为747.21μmol/L.实验数据表明,爪哇黄芩5,6,2'-三羟基-7-甲氧基二氢黄酮为爪哇黄芩活性二氢黄酮单体成分.
To study the anti - tumor activities effects of flavanone in Scutellaria javanica Jungh. , active fraction and sensitive cell line were selected. The effect on HepG - 2 cell proliferation was researched to reveal the anti - tumor activities of flavanone in Scutellaria ja-vanica Jungh. This paper provided the experimental foundation for develop natural drugs.The human liver cancer cell HepG - 2, gastric cancer cell SGC - 7901, lung cancer cell A549, breast cancer cell MCF -7 and C6 glioma cells, cultivated in vitro, were used as models in the paper. With median inhibitory concentration (IC50) used as index,MTT col-orimetry was applied to ascertain the anti - proliferative effects of petroleum ether, dichlo- romethane and n - butyl alcohol extraction fractions in Scutellaria javanica Jungh. on the a-bove - mentioned 5 cancer cells. The dichloromethane extracted fraction of Scutellaria javan-ica Jungh. was its active fraction. HepG -2 cells is its sensitive cell line. The MTT colorimetry was adopted to ascertain the inhibiting effects of flavanone monomers extracted from the active fractions on human liver cancer cell HepG -2 to screen active monomers. The results suggested that the 5,6,2r - trihydroxy - 7 - methoxy flavanone IC50 of Scutellaria javanica Jungh. was 110. 3 μmol/L, and the 5 - hydroxyl -6,7,2,,6, - tetramethyl flavanone IC50 was 747.21 μmol/L. It was discovered that the 5 , 6 , 2' - trihydroxy - 7 - methoxy flavanone was the active composition of Scutellaria javanica Jungh. on human liver cancer cell HepG - 2 .
