Study on social behavior and abilities of learning and memory in TS mice

摘要 对二丙腈诱导抽动症(TS)小鼠社交与学习记忆能力进行考察,测定脑组织多巴胺(DA)水平初步探讨其机制.60只ICR小鼠按0 d社交成绩随机分为对照组(n=20)与模型组(n=40),腹腔注射亚氨基二丙腈(IDPN)诱导TS模型,于0、10、20、30 d测定自主活动、点头次数、攀爬时间,0、15、30d检测社交能力,30 d Morris水迷宫检测学习记忆能力,实验结束后断头取脑,检测脑组织多巴胺质量浓度.结果表明,与对照组相比,模型建立10 d后,点头次数、攀爬时间有明显差异(P<0.05),20~30 d,模型组自主活动、点头次数增加、攀爬时间减少,均有显著变化(P<0.01);15 d模型组社交次数、时间减少(P<0.05)并延续至30 d;小鼠水迷宫潜伏期无明显变化(P>0.05);模型组小鼠脑组织DA质量浓度增加(P<0.05).TS模型伴随社交障碍,但与学习记忆能力不相关,并伴随DA质量浓度增加.为今后TS深入研究与药物治疗提供依据. In this paper, the social behavior and the abilities of learning and memory in TS mice were investigated,and the DA level in brain tissue was measured and then the mecha-nism of it was discussed. A total of 60 ICR mice were randomly divided into control group ( n = 20) and model group ( n = 40) based on 0 d social score. Intraperitoneal inject IDPN to establish TS model,spontaneous activities, nodding movements, climbing behavior were measured in 0,10, 20 and 30 d. In 0,15 and 30 d,the social scales were detected, and the learning and memories were measured in 30 d by Morris water maze. At the end of the experiment, the brain tissue was taken to detect the DA level. Results indicated that com-pared to the control group, after 10 d successfully established the model, nodding movements and climbing behavior were significantly different ( P 〈 0.05 ) , and after 20 d to 30d, spon-taneous activity, nodding movements in model group were significantly decreased ( P 〈0. 01) . The number and time of social behavior were decreased and extend to 30 d (P 〈0. 05 ) . The escape latency of Morris water maze have no significant changes (P 〉0. 05) , and the DA content in mice brain was increased (P 〈0. 05) . TS model may occur social obstacle ,but there were no evidence showed that it had relevance with learning and memories, and along with the increase of DA content. This investigation could provide evidence to deep-ly develop and drug treated.
