过表达含Ⅰ型血小板结合蛋白基序的去整合素样金属蛋白酶18基因抑制人骨肉瘤细胞的增殖与迁移 被引量:3

摘要 目的探讨含Ⅰ型血小板结合蛋白基序的去整合素样金属蛋白酶(ADAMTS)18基因对人骨肉瘤细胞增殖与迁移作用及其机制。方法将ADAMTS18过表达载体和空载体分别转染143B和U-20S人骨肉瘤细胞,细胞计数试剂盒(CCK)-8和Transwell小室实验研究过表达ADAMTS18基因对143B和U-20S细胞增殖和迁移作用,蛋白质Western印迹实验检测ADAMTS18和蛋白激酶B(AKT)蛋白表达水平。结果与空载体转染细胞株比较,过表达ADAMTS18基因可显著抑制143B和U-20S细胞增殖和迁移能力,并下调143B和U-20S细胞AKT蛋白水平。结论 ADAMTS18经下调AKT通路活性而抑制人骨肉瘤细胞恶性表型。
