摘要
目的:观察金刚健骨片对骨质疏松症模型大鼠体内OPG/RANKL/RANK系统表达的影响。方法:采用卵巢摘除法建立SD大鼠骨质疏松模型,设立假手术组、模型组(单纯去卵巢组)、雌激素组(尼尔雌醇组)和金刚健骨片组。造模成功后4周开始给药,给药12周后RT-PCR检测各组大鼠骨组织中OPG/RANKL/RANK mRNA的表达。结果:与假手术组比较,模型组的OPG表达降低、RANKL表达增加,差异具有统计学意义(P<0.05);与模型组比较,金刚健骨片组和雌激素组的OPG表达增高,RANKL表达降低,差异均有统计学意义(P<0.05);金刚健骨片组与雌激素组之间比较,差异无统计学意义(P>0.05);RANK的mRNA表达在各组之间比较,差异无统计学意义(P>0.05)。结论:采用卵巢摘除法成功建立大鼠骨质疏松模型;金刚健骨片可促进OPG在骨质疏松大鼠中的表达,抑制其RANKL的表达,这可能是金刚健骨片治疗骨质疏松症的作用机制之一。
Objective: To investigate the effect of Jingang Jiangu tablets on the OPG/RANKL/RANK system in a rat model of osteoporosis. Methods: Sprague-Dawley rats were ovariectomized to establish a model of osteoporosis and then divided into sham-operation group,model group( ovariectomization alone),estrogen group( nilestriol),and Jingang Jiangu tablet group. The drugs were administered since four weeks after the model was established,and RT-PCR was used to measure the mRNA expression of OPG/RANKL/RANK in bone tissue after 12 weeks of drug administration. Results: Compared with the sham-operation group,the model group had a significant reduction in the expression of OPG and a significant increase in the expression of RANKL( P 0. 05); compared with the model group,the Jingang Jiangu tablet group and the estrogen group had a significant increase in the expression of OPG and a significant reduction in the expression of RANKL( P 0. 05); there were no significant differences between the Jingang Jiangu tablet group and the estrogen group( P 0. 05); there was no significant difference in the mRNA expression of RANK between these groups( P 0. 05). Conclusion: A rat model of osteoporosis is successfully established by ovariectomization. Jingang Jiangu tablets can promote the expression of OPG and inhibit the expression of RANKL in rats with osteoporosis,which may be one of the mechanisms of action of Jingang Jiangu tablets in the treatment of osteoporosis.
