响应曲面法优化苦丁茶总皂苷的提取工艺 被引量:3

Optimization of the extraction process for total saponins in Kudingcha by response surface methodology

摘要 目的:采用Box-Behnken响应曲面设计法确定苦丁茶总皂苷的最佳醇提工艺。方法:本研究在单因素实验的基础上,以药食同源中药苦丁茶中主要皂苷成分苦丁茶皂苷A(kudinoside A)为对照品,通过响应曲面法对影响苦丁茶中总皂苷得率的不同因素进行优化,以期研究乙醇提取苦丁茶总皂苷的工艺条件。结果:最佳提取工艺条件为加入乙醇浓度63%,料液比1∶16,提取时间90min。实验结果所得苦丁茶总皂苷得率为5.59%。结论:本实验结果与预测值高度拟合,为进一步开发和利用苦丁茶资源以及新药研发提供了科学依据。 Objective: To investigate the optimal ethanol extraction process for total saponins in Kudingcha by Box-Behnken response surface methodology. Methods: On the basis of single-factor experiment,and with kudinoside A,a major saponin constituent in Kudingcha for both food and medicine,as reference,the response surface methodology was used to optimize the factors which influenced the yield of total saponins in Kudingcha,in order to obtain the optimal process conditions for ethanol extraction of total saponins in Kudingcha. Results: The optimal process conditions were an ethanol concentration of 63%,a solid-liquid ratio of 1∶ 16,and an extraction time of 90 minutes. The yield of total saponins in Kudingcha was 5. 59%. Conclusion: The results of this experiment are highly fitted to predictive values and thus provide a scientific basis for further exploitation and utilization of Kudingcha resources and research and development of new drugs.
