miR-125b通过靶向STAT3抑制胃癌细胞SGC-7901的侵袭迁移能力

miR-125b inhibits the invasion and migration of gastric cancer cell SGC-7901 by targeting STAT3

目的研究miR-125b靶向STAT3对胃癌细胞SGC-7901侵袭迁移能力的影响。方法运用qPCR法检测miR-125b在胃癌及癌旁组织中的表达,运用Western blot法检测STAT3在胃癌组织及癌旁正常组织中的表达,用双荧光素酶报告基因系统检测miR-125b对STAT3转录活性的影响,用Transwell实验检测过表达miR-125b对SGC-7901细胞侵袭迁移能力的影响,用Western blot法检测过表达miR-125b后SGC-7901细胞STAT3的表达变化。结果与癌旁正常组织比较,胃癌组织miR-125b的表达水平明显降低,STAT3蛋白表达水平明显升(P<0.01)。双荧光素酶报告基因系统检测结果显示,miR-125b可以直接调控STAT3的转录活性。过表达miR-125b后,SGC-7901细胞的侵袭和转移能力明显降低,STAT3的表达水平下调(P<0.01)。结论miR-125b可靶向STAT3的表达调控SGC-7901细胞的侵袭迁移能力。

Objective To investigate the effect of miR-125b targeting STAT3 on the invasion and migration of gastric cancer cells 5GC-7901. Methods The expression of miR-125b in gastric cancer and adjacent tissues was detected by qPCR method. The expression of STAT3 in gastric cancer and adjacent tissues was detected by Western blot method. The effect of miR-125b on the transcriptional activity of STAT3 by double luciferase reporter gene system. The Transwell test was used to detect the ability of invasion and migration of SGC-7901 cells after overexpression of miR-125b. The expression of STAT3 in SGC-7901 cells was detected by Western blot after overexpression of miR-125b. Results Compared with the normal tissues, the expression level of miR-125b in gastric cancer tissue was significantly decreased, but increased significantly for the expression of STAT3 protein（P〈0.01）. The double luciferase reporter gene system showed that miR-125b could directly regulate the transcriptional activity of STAT3. The overexpression of miR-125b decreased the invasion and metastasis of SGC-7901 cells significantly, and down-regulated the expression level of STAT3（P〈0.01）. Conclusion miR-125b could regulate the invasion and migration of SGC-7901 cells by targeting STAT3.
