Th9及相关细胞因子IL-9在重症肌无力患者外周血中的表达及意义

Expression and significance of Th9 and related cytokines IL-9 in peripheral blood of patients with myasthenia gravis

摘要 目的 探讨Th9细胞及相关细胞因子白细胞介素-9(IL-9)在重症肌无力(MG)患者外周血中的表达及其临床意义.方法 选取明确诊断的MG患者39例(MG组)及同期健康对照者30例(对照组).抽取外周静脉血,采用流式细胞术检测外周血Th9细胞比例,ELISA检测血清IL-9浓度,放免法检测抗乙酰胆碱受体抗体(anti-AchR)浓度,Spearman分析各指标间的相关性.结果 MG组外周血Th9细胞比例高于对照组[(1.07±0.23)%比(0.27±0.11)%,P<0.05],血清IL-9水平高于对照组[(4.73±1.87) pg/ml比(1.55±0.51) pg/ml,P<0.05].GM患者外周血Th9细胞比例与血清IL-9浓度呈显著正相关(r=2.658,P<0.05),与Anti-AchR无显著相关性(r =0.911,P>0.05),而IL-9与Anti-AchR浓度间呈正相关(r=2.131,P<0.05).结论 Th9细胞及其效应细胞因子IL-9在MG患者外周血中异常上调,提示其可能参与了MG的发生发展过程. Objective To investigate the expression and clinical significance of Th9 cell and its effector cytokine interleukin-9 (IL-9) in peripheral blood of patients with myasthenia gravis (MG).Methods Thirty-nine patients diagnosed with MG (MG group) were selected and 30 healthy people (control group) during the same period were chosen.The percentage of Th9 cell in peripheral blood was detected by flow cytometry,serum IL-9 concentration was determined by ELISA,and anti-AchR was measured by radioimmunoassay.The correlations among each index were analyzed with Spearman.Results The proportion of Th9 cells in peripheral blood of MG group was higher than that of control group [(1.07 ± 0.23) % vs.(0.27 ± 0.11) %,P 〈 0.05],and serum IL-9 level was higher than that in control group [(4.73 ± 1.87) pg/ml vs.(1.55 ± 0.51) pg/ml,P 〈 0.05].The proportion of Th9 in peripheral blood of MG patients was positively correlated with serum IL-9 concentration (r =2.658,P 〈0.05),and was not correlated with anti-AchR(r =0.911,P 〉 0.05).However,IL-9 and Anti-AchR concentrations were positively correlated (r =2.131,P 〈 0.05).Conclusions Th9 cells and the effector cytokine IL-9 are abnormally up-regulated in peripheral blood of patients with MG,it is suggested that they may be involved in the pathogenic mechanism of MG.
