肺表面活性物质参与尼古丁诱导支气管上皮细胞高迁移率族蛋白1释放 被引量:1

Nicotine induced expression of high-mobility group box 1 via surfactant proteins A in human airway epithelial cells

摘要 目的 本研究旨在探讨支气管上皮细胞肺表面活性物质蛋白 (SP-A)在尼古丁诱导支气管上皮细胞高迁移率族蛋白1 (HMGB1)释放中的作用。方法 尼古丁 (6×10-6mol/L)刺激支气管上皮细胞株16HBE,采用酶联免疫吸附测定 (ELISA)方法检测刺激6h、12h、24h后细胞培养上清中 HMGB1浓度。预先加入SP-AsiRAN孵育1h,再加入尼古丁刺激16HBE24h,ELISA方法检测 HMGB1的分泌。结果 尼古丁刺激24h后 HMGB1的分泌量较未给予尼古丁刺激的正常对照组明显增加,刺激6h和12h后分泌量较正常对照组稍增加,但分泌量差异无统计学意义。加入SP-AsiRAN后尼古丁刺激24h组,HMGB1的分泌量显著增加。结论 SP-A参与了尼古丁诱导支气管上皮细胞 HMGB1释放。 Objective To test whether nicotine induces the expression of high mobility group box 1 (HMGB1) in human bronchial epithelial cells (16HBECs) in vitro via surfactant proteins A (SP-A). Methods 16HBECs were exposed to nicotine (6× 10 -6 μmol/L) for 6 h, 12 h and 24 h, respectively. The secretion of HMGB1 was assessed by enzyme-linked immunosorbent assay (ELISA). Results We showed that ceils exposed to nicotine for 24 h exhibited significantly increased HMGB1 secretion, Nicotine treatment had no significant effect on the secretion of HMGB1, although a slight upregulation of HMGB1 secretion was observed at 6 h and 12 h. Knockdown of SP-A with small interfering RNA (siRNA) promoted the secretion of tlMGB1. Conclusions These results suggest that nicotine can induce the expression of HMGB1 via SP-A in HBECs.
