摘要
[目的]优化纹党组织培养的培养基成分,探索高效的愈伤组织诱导及植株再生技术。[方法]以文县纹党为材料,研究外植体不同部位,不同浓度激素配比培养基及外植体不同部位消毒时间对纹党愈伤组织诱导及植株再生的影响。[结果]带芽侧茎出愈率和愈伤组织分化效果最好,叶片处理效果最不理想。带芽侧茎愈伤组织在激素浓度配比为2,4-D 0.5 mg/L+6-BA 1.0 mg/L的MS培养基上诱导率可达95%,而后在相同浓度的激素配比下继代培养2~3次后,在激素浓度配比为6-BA 0.5 mg/L+NAA 1.0 mg/L的MS培养基上诱导再分化出芽率也可达95%。[结论]通过组织培养初步建立了文县纹党愈伤组织的快速繁殖方法 ,为这一地域特色鲜明的药用植物资源的快速繁殖及扩大培养奠定了理论基础。
[Objective] This study was to optimize medium component of Codonopsis pilosula (Franch.) Nannf 'Wenxian' ,in order to explore efficient technology of callus induction and plant regeneration. [Method] This paper studied the effects of different explants, different concentrations of hormones on callus induction and plant regeneration of Codonopsis pilosula (Franch.) Nannf ' Wenxian'. [Result] The callus and callus differentiation effect of stem with buds was best. The callus induction of stem with buds was 95% on the MS medium with 2,4-D 0.5 mg/L + 6-BA 1.0 mg/L. After 2-3 generations of subclture, the rate of re-emergence budding was up to 95% on MS medium with 6-BA 0.5 mg/L + NAA1.0 mg/L. [Conclusion] The rapid propagation method of Wenxianmao callus was established initially. It provided a theoretical foundation for further study of this medicinal plant resources.
