四逆散对创伤后应激障碍大鼠海马神经元信号强度的影响 被引量:6

Effect of Sinisan on hippocampal neurons signal intensity in rats with post-traumatic stress disorder

摘要 目的研究四逆散对大鼠海马CA1/CA3区神经元时空特性的影响。方法按照随机数表法将SD大鼠随机分为5组,每组10只:正常组、空白组、模型组、对照组和实验组。除正常组、空白组不造模型外,其余各组用电流刺激加黑暗幽闭禁锢法复制大鼠创伤后应激障碍(PTSD)模型。空白组与模型组于造模前1 h均给予0.9%NaCl 10 mL·kg^(-1)。对照组和实验组分别灌胃给予盐酸帕罗西汀4.2 mg·kg^(-1)和四逆散2.41 g·kg^(-1)。用在体多通道神经信号采集系统采集信号,记录大鼠海马的CA1、CA3区100 Hz内的动作电位信号频谱,从Neuro Explorer神经数据分析软件导出,生成功率谱密度频谱图。结果与正常组大鼠海马CA1区的神经元动作电位谱功率集中分布在(-109.08±15.41)d B、CA3区的分布在(^(-1)08.46±13.16)d B比较,空白组分别是(-110.17±14.30),(-109.54±14.16)d B,差异均无统计学意义(均P>0.05);造模后大鼠海马区各频段功率谱密度分别是(-117.39±14.97),(-122.51±16.42)d B,模型组与空白组比较,差异均有统计学意义(均P<0.01)。给药后,实验组大鼠海马区各频段功率谱密度分别是(-110.83±11.40),(-110.36±10.87)d B,与模型组比较,差异均有统计学意义(均P<0.01)。结论四逆散对PTSD大鼠海马区神经元信号强度有明显的正向调节作用。 Objective To study the influence of Sinisan on hippocampal CA1 and CA3 neuron spatiotemporal characteristics in rats with post - traumatic stress disorder(FFSD). Methods SD rats were divided into five groups: normal group, blank group, model group, control group, experimental group. Except for normal group and blank group, the rats in other groups were stimulated by the current stimulation plus dark claus- trophobic method to replicate the posttraumatic stress disorder (PTSD) model. The blank group and model group were given 0. 9% NaC1 l0 mg·kg-1 at 1 h before making the model. The control group and experi- mental group was given paroxetine hydrochloride 0.42 mg·kg-1 and Sinisan( containing crude drug 0.24 g · mL-1)by intragastric administra- tion,respectively. The model replicated 7 d, and each electric shock was 30 rain every morning and afternoon. The neural coding on amplitude in hippocampal CA1 and CA3 were detected by the M -NEMEA after ad- ministration, and the spectral density spectrum was obtained from Neuroexplorer neural data analysis software. Results The power spectral densities of CA1 and CA3 in hippocampus of rats were determined, they were( - 109.08 ± 15.41 ) and( - 108.46 ± 13.16) dB in normal group, ( - 110. 17 ± 14.30) and ( -109.54 ±14. 16)dB in blank group, and the differences were not statistically significant (all P 〉0.05). Compared with the blank group, the power spectral densities of hippocampus in the model group decreased significant- ly, which were ( - 117.39 ± 14. 97 ), ( - 122. 51 ± 16.42 ) dB, and the differences were statistically significant ( all P 〈0. 01). Compared with the model group, the power spectral densities of the experimental group in hippoeampus were significantly higher ( P 〈 0. 01 ), which were ( - 110. 83 ± 11.40), ( - 110. 36 ± 10. 87 ) dB. Conclusion The Sinisanhas significant positive effect on hippoeampal neuron signal intensity.
