摘要
目的:探讨通关藤苷元甲(tenacigenin A)对肝细胞癌小鼠肝功能的保护作用及其可能机制。方法:建立二乙基亚硝胺(DEN,2 mg·kg^(-1))联合四氯化碳(CCl4,1 595 mg·kg^(-1))诱导的雌性小鼠肝细胞癌模型,受试组小鼠分别灌胃给予通关藤苷元甲(10、20 mg·kg^(-1)),每周6天,共4周。计算肿瘤结节数,微板法检测各组血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、碱性磷酸酶(AKP)水平;Elisa法检测血清中甲胎蛋白(AFP)含量;HE染色法观察肝脏病理学改变;Western Blot检测肝组织中Bax、Bcl-2水平。结果:通关藤苷元甲(10,20 mg·kg^(-1))组小鼠的肝、脾指数及血清中AST、ALT、AKP、AFP和Bcl-2水平较模型组均显著降低,Bax则显著升高(P<0.05);肝脏的病理学损伤情况有所减轻。结论:通关藤苷元甲对肝细胞癌雌性小鼠的肝功能具有一定的保护作用,其机制可能与其促进肝癌细胞凋亡等有关。
Objective: To investigate the protective effects of tenacigenin A on liver function of mice with hepatocellular carcinoma (HCC) and the underlying mechanism. Methods: Diethylnitrosamine (DEN, 2 mg·kg-1) combined with CCI4 (1 595 mg·kg-1) were used to establish a hepatocellular carcinoma mouse model and the experimented groups were intragastrically administrated with te- nacigenin A ( 10 or 20 mg ·kg-1 ) for 4 weeks (6 days every week). The number of nodules was calculated. The serum activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkalinephosphatase (AKP) were determined by microplate test and alpha fetoprotein (AFP) was analyzed by Elisa kits. Hematoxylin-Eosin (HE) staining was used to evaluate the hepatic patho- logical lesions. Simultaneously, the protein levels of Bax and Bcl-2 were analyzed by western blot. Results: Tenacigenin A could re- duce the liver and spleen indices and the serum levels of AST, ALT, AKP, AFP and Bcl-2, and increase the level of Bax, meanwhile, it improved the hepatic pathological lesions. Conclusion: The results suggested that tenacigenin A shows a good protective action on liver function of female mice with HCC, and the effects may be associated with the apoptosis of hepatoma cells induced by tenacigenin A.
