基于SSR标记的黄精品种(系)DNA指纹图谱库构建 被引量:20

Construction of DNA Fingerprint Database Based on SSR Marker for Polygonatum Varieties(Lines)

摘要 为了开展药用黄精种质资源的分子标记育种工作,实现对黄精品种及优良种质的快速精准鉴定。本研究采用12对引物对32个野生黄精种质材料进行SSR多态位点分析,之后计算相似系数并进行聚类分析,然后构建DNA指纹图谱及其QR编码。结果表明:12对引物共扩增出127条条带,其中多样性条带125条,多态率为98.43%,每对核心引物检测到的多样性条带数为3~17条,平均每对为10.42条,PIC(多样性信息含量)平均为0.46。其中等位基因数为1.984 3,有效等位基因数为1.527 8,Nei's基因多样性为0.314 9,Shannon's指数为0.478 8。SSR聚类结果很好的反映供试黄精材料的亲缘关系,所构建的DNA指纹图谱能对所有的种质材料进行高效区分。QR编码高效、方便的录入大量信息,有利于黄精品种及优良种质的鉴定工作。该体系的建立为黄精种质资源遗传多样性分析及品种保护、优良品种选育、优良种质鉴定提供依据。 The purpose of the study was to develop the molecular marker breeding of medicinal Polygonatum germplasms, and to achieve rapid and accurate identification of Polygonatum varieties and their fine germplasms. It has adopted 12 pairs of primers to analyze SSR polymorphism of 32 wild Polygonatum germplasms. Then the similarity coefficient calculation and clustering analysis based on UPGMA algorithm were carried out, and the DNA fingerprint and QR coding system were constructed. The results of the study showed that 127 bands were amplified from the 12 pairs of primers, among which 125 were polymorphic bands, with a polymorphism rate of 98.43%. Each pair of primers could be detected with 3-17 bands of polymorphic bands, with an average of 10.42. The average content of polymorphic information was 0.46. In addition, the number of alleles was 1.984 3, the number of effective alleles was 1.527 8, Nei's gene diversity was 0.314 9, and Shannon's index was 0.478 8. The SSR clustering results presented a good reflection of the genetic relationship among the tested Polygonatum materials, and the constructed DNA fingerprints helped efficiently differentiate all the germplasm materials. The QR coding, which stored large quantities of information in a high efficient way, was conductive to the identification of fine Polygonatum varieties and germplasms. Therefore, the establishment of the system might provide the basis for genetic diversity analysis and variety protection, breeding of fine varieties, and the identification of fine Polygonatum germplasms.
