摘要
高分辨率熔解曲线(HRM)技术是一种通过饱和荧光染料实时监测双链DNA的熔解过程,以此反映扩增子DNA序列特性的基因分析新技术。该技术具有快速、高效、高特异性、污染小、廉价等优点,目前已广泛应用于临床医学、生命科学、法医学、药材和食品鉴定领域。本文对高分辨率熔解曲线技术及其在临床医学的应用进行综述。
High-resolution melting(HRM) is a new technology of new gene analysis based on monitoring double-stranded DNA melting process with saturated fluorescent dyes in real time to reflect the amplicon DNA sequence characteristics.The technology has the advantages of fast,high efficiency,high specificity,low pollution,and low cost.Therefore,it has been widely used in clinical medicine,bioscience,forensic medicine,herbs and food identification and other fields.This paper reviews the basic principles,key factors,and clinical application of HRM.
