摘要 目的探索雷公藤甲素对于人耐药乳腺癌细胞的抑制作用及其对阿霉素化疗敏感性的影响。方法体外培养阿霉素耐药乳腺癌细胞MCF-7/ADR细胞株,采用不同浓度的雷公藤甲素和阿霉素单药或联合用药处理。利用四甲基偶氮唑盐比色法(MTT法)检测各组细胞的增殖活性,利用流式细胞术检测各组细胞凋亡及细胞周期。结果雷公藤甲素可抑制阿霉素耐药乳腺癌癌细胞的增殖,且存在着明显的剂量-效应关系。与单独使用雷公藤甲素或者阿霉素相比,雷公藤甲素联合阿霉素组的细胞凋亡率明显增高。结论雷公藤甲素不仅能对耐药乳腺癌细胞的增殖产生抑制作用,同时也可以增强乳腺癌细胞对阿霉素化疗的敏感性。 Objective To determine the inhibitory effect of triptolide on human breast cancer cells and its mechanism. Methods Human breast cancer cell lines were cultured in vitro and treated with various concentrations of triptolide and/or adriamycin. MTT assay was used to detect the proliferation of breast cancer cells in vitro. Flow cytometry was used to analyze the apoptosis and cell cycle. Results Triptolide inhibited the proliferation of adriamycin-resistant breast cancer cells with significant dose-effect relationship. Triptolide combined with adriamycin significantly induced the apoptosis of MCF-7/ADR cells, much higher than the use of triptolide or adriamycin alone. Conclusion Triptolide can inhibit the proliferation of breast cancer cell lines, and enhance the sensitivity of adriamycin to breast cancer chemotherapy.
