和降颗粒乙醇提取工艺研究及初步药效学评价 被引量:4

Optimization of ethanol-extraction for Hejiang granules and its pharmacodynamics

摘要 目的优选和降颗粒的乙醇提取工艺条件,并与传统汤剂及配方颗粒进行初步药效学对比研究,对提取工艺进行评价,为该方进一步开发提供参考。方法以黄连总生物碱提取率、浙贝生物碱提取量、干膏得率的综合评分为指标,采用正交试验法,优选提取工艺条件。采用酶联免疫吸附法(ELISA)测定基础致敏结合食管酸灌注所致NERD大鼠模型中相关蛋白含量,对和降颗粒的药效进行评价,并对量效关系进行考察。结果最佳提取工艺为黄连、浙贝母采用12倍50%乙醇(考虑药材对溶剂的吸收首次加液量为13.4倍)、提取3次、每次1.5 h。药效研究表明和降颗粒低剂量组药效显著,日服药剂量降为原来的1/2。量效关系考察发现,和降颗粒中黄连总生物碱含量高于传统汤剂及配方颗粒,量效关系明显。结论该工艺稳定可行,保证了和胃降逆方药效物质的充分提取,大大降低了服用剂量,为进一步开发利用该处方打下了坚实基础。 Objective To optimize the alcohol-extraction process of Hejiang granule, and to evaluate the extraction process and provide reference for further development of this prescription by comparative preliminary study of pharmacodynamics between Hejiang granule, traditional decoction and formula granules. Methods Orthogonal test was used to optimize the ethanol-extraction process, and the comprehensive score of coptis alkaloids extraction rate, fritillaria alkaloids extraction amount and dry paste yield was used as indexes. ELISA was used to evaluate the efficacy by testing related protein of the NERD rat model induced by basic sensitization combined with esophageal acid infusion. And dose-effect relationship was investigated. Results The best extraction process was that coptis and fritillaria were extracted with 12 folds of 50% ethanol for 3 times (considering the absorption of the solvent, so the first time solvent adding quantity was 13.4 times), and 1.5 hours each time. The total content of epiberberine, coptisine chloride, palmatin hydrochloride and berberine hydrochloride in this granule was 13.3 mg · g -1. The pharmacodynamics study showed that the low dose group of Hejiang granule was better than that of the high dose group of traditional decoction and low dose group of formula granules with significant difference. Accordingly, the daily dose dropped to 1/2 for the original dosage. Conclusion The process is not only stable and feasible, but also ensures the extraction of effective substances from Heweijiangni prescription, and greatly reduces the dosage, laying a solid foundation for further development and utilization of the prescription.
